Abstract
Keberhasilan dalam sektor budidaya perikanan sangat dipengaruhi oleh berbagai aspek diantaranya kualitas benih ikan yang digunakan, sistem budidaya, lalu lintas/peredaran perdagangan ikan, kualitas lingkungan sekitar kawasan budidaya, peran serta atau aksesbilitas pembudidayaan ikan/udang ke institusi serta teknik pengendalian penyakit ikan. Penyakit yang disebabkan oleh bakteri seperti contoh bakteri V. harveyi. Bakteri ini dapat menularkan penyakit pada biota yang dibudidayakan. Salah satu alternatif yang digunakan untuk mengatasi permasalahan penyakit tersebut yaitu dengan cara menggunakan antibiotik dari bahan alami daun nangka. Daun nangka mengandung antibakteri yaitu alkaloid, saponin, falvonoid, dan tanin. Penelitian dilakukan di Laboratorium Budidaya Ikan Divisi Penyakit dan Kesehatan Ikan, Fakultas Perikanan dan Ilmu Kelautan, Universitas Brawijaya, Malang. Penelitian dilaksanakan pada bulan Februari 2021 - April 2021. Tujuan penelitian ini adalah untuk mengetahui penggunaan ekstrak daun nangka sebagai antibakteri terhadap bakteri V. harveyi. Metode penelitian yang digunakan yaitu metode eksperimen. Metode eksperimen adalah metode untuk mencari pengaruh perlakuan tertentu dalam kondisi yang terkendali. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) menggunakan 5 perlakuan dan 3 ulangan. Dosis perlakuan ekstrak daun nangka yaitu A (50 ppm), B (100 ppm), C (150 ppm), D (200 ppm), dan E (250 ppm). Hasil ini ditunjukkan adanya zona bening pada sekitar kertas cakram akibat dosis perlakuan yang diberikan. Dosis yang menghasilkan ukuran diameter zona bening tertinggi didapatkan dari perlakuan E (250 ppm) dengan rerata zona hambat 8,95 ± 0,90 mm. Dosis yang menghasilkan ukuran diameter zona bening terendah didapatkan dari perlakuan A (50 ppm) dengan rerata zona hambat 7,11 ± 0,19 mm. Hubungan antara dosis ekstrak daun nangka dengan ukuran diameter zona hambat menghasilkan kurva linier dengan persamaan y = 0,0012x + 7,0187 dan koefisien determinasi (R2) sebesar 0,7077. Kurva ini menunjukkan bahwa dosis perlakuan yang diberikan mengalami peningkatan pada zona hambat. Berdasarkan hasil penelitian yang telah dilakukan dapat ditarik kesimpulan bahwa ekstrak kasar daun nangka memiliki manfaat sebagai bioaktif alami terhadap bakteri V. harveyi. Ekstrak daun nangka memiliki sifat bakteriostatik atau bersifat menghambat pertumbuhan, hal ini ditunjukkan dari ukuran diameter zona hambat yang mengalami penurunan dari inkubasi 24 jam ke inkubasi 48 jam. Zona hambat yang tertinggi yaitu diperoleh pada dosis 250 ppm sebesar 8,95 ± 0,90 dan yang terendah terdapat pada dosis 50 ppm sebesar 7, 11 ± 0,19.
English Abstract
Success in the aquaculture sector is strongly influenced by various aspects including the quality of fish seeds used, aquaculture systems, fish trade traffic/circulation, the quality of the environment around the cultivation area, participation or accessibility of fish/shrimp farming to institutions and fish disease control techniques. Diseases caused by bacteria such as the example of the bacterium V. harveyi. These bacteria can transmit diseases to cultivated biota. One alternative that is used to overcome the problem of the disease is by using antibiotics from natural ingredients of jackfruit leaves. Jackfruit leaves contain antibacterial, namely alkaloids, saponins, flavonoids, and tannins. The research was conducted at the Fish Cultivation Laboratory, Fish Disease and Health Division, Faculty of Fisheries and Marine Sciences, Universitas Brawijaya, Malang. The study was conducted in February 2021 - April 2021. The purpose of this study was to determine the use of jackfruit leaf extract as an antibacterial against V. harveyi bacteria. The research method used is the experimental method. The experimental method is a method to find the effect of certain treatments under controlled conditions. The research design used was a completely randomized design (CRD) using 5 treatments and 3 replications. The treatment doses of jackfruit leaf extract were A (50 ppm), B (100 ppm), C (150 ppm), D (200 ppm), and E (250 ppm). This result is indicated by the presence of a clear zone around the paper disc due to the treatment dose given. The dose that produced the highest clear zone diameter was obtained from treatment E (250 ppm) with an average inhibition zone of 8.95 ± 0.90 mm. The dose that produced the lowest clear zone diameter was obtained from treatment A (50 ppm) with an average inhibition zone of 7.11 ± 0.19 mm. The relationship between the dose of jackfruit leaf extract and the diameter of the inhibition zone resulted in a linear curve with the equation y = 0.0012x + 7.0187 and the coefficient of determination (R2) was 0.7077. This curve shows that the treatment dose given has increased in the inhibition zone. Based on the results of the research that has been done, it can be concluded that the crude extract of jackfruit leaves has benefits as a natural bioactive against V. harveyi bacteria. Jackfruit leaf extract has bacteriostatic or growth-inhibiting properties, this is indicated by the diameter of the inhibition zone which decreased from 24-hour incubation to 48-hour incubation. The highest inhibition zone was obtained at a dose of 250 ppm of 8.95 ± 0.90 and the lowest was found at a dose of 50 ppm of 7.11 ± 0.19.
