Abstract

Karies gigi adalah penyakit yang merusak struktur gigi yang ditandai dengan adanya demineralisasi jaringan keras gigi yang diakibatkan oleh akumulasi bakteri Streptococcus mutans sebagai agen utama pemicu terjadinya karies menurut beberapa hasil penelitian. Tumbuhan yang memiliki kandungan sebagai antibakteri banyak ditemui dan diteliti di Indonesia salah satunya adalah kopi Robusta (Coffea cenephora). Beberapa komponen dalam biji kopi Robusta yang memiliki aktifitas sebagai antibakteri yaitu kafein, trigonelin dan asam klorogenat. Tujuan dari penelitian ini untuk mengetahui efektivitas seduhan biji kopi Robusta (Coffea canephora) terhadap pertumbuhan bakteri Streptococcus mutans secara in vitro. Metode yang digunakan yaitu dilusi tabung untuk menentukan Kadar Hambat Minimal (KHM) dan Kadar Bunuh Minimal (KBM). Konsentrasi seduhan yang digunakan yaitu 14gr/dl, 17gr/dl, 20gr/dl, 23gr/dl, dan 26gr/dl. Hasil penelitian KHM tidak dapat dihitung karena warna cokelat kehitaman dari seduhan kopi sehingga tidak dapat dilihat kekeruhannya, sedangkan untuk KBM didapatkan hasil sebesar 20gr/dl. Hasil analisa data menggunakan perhitungan uji statistik non-parametrik Kruskal-Wallis dan hasil uji korelasi Spearman menunjukkan adanya hubungan bermakna yang sangat kuat dari peningkatan konsentrasi seduhan biji kopi Robusta terhadap pertumbuhan bakteri Streptococcus mutans. Kesimpulan dari penelitian ini adalah efektivitas seduhan biji kopi Robusta (Coffea cenephora) dalam menghambat pertumbuhan bakteri Streptococcus mutans secara in vitro dengan metode dilusi tidak dapat memberikan hasil yang signifikan namun dari hasil data menunjukkan penurunan pertumbuhan jumlah koloni bakteri Streptococcus mutans.

English Abstract

Dental caries is a disease that damages the structure of the teeth which is characterized by demineralization of tooth caused by the accumulation of plaque bacteria Streptococcus mutans as the main agent that triggers caries according to several studies. Plants that contain antibacterial properties are found and researched in Indonesia, one of which is Robusta coffee (Coffea robusta). Several components in Robusta coffee beans that have antibacterial activity are caffeine, trigonelline, and chlorogenic acid. The purpose of this study was to determine the effectiveness of steeping Robusta coffee beans (Coffea canephora) on the growth of Streptococcus mutans bacteria in vitro. The method used is tube dilution to determine the Minimum Inhibitory Concentration (MIC) and the Minimum Bactericidal Concentration (MBC). The concentration of the brew used was 14gr / dl, 17gr / dl, 20gr / dl, 23gr / dl, and 26gr / dl. The results of the MIC could not be calculated because of the blackish-brown color of the coffee steeping so that the turbidity could not be seen, while for the MBC the results were 20gr / dl. The results of data analysis used the non-parametric statistical test calculation Kruskal-Wallis and the Spearman correlation test show that there was a very strong significant relationship between the increase in the concentration of steeping Robusta coffee beans on the growth of Streptococcus mutans. The conclusion of this study is the effectiveness of steeping Robusta coffee beans (Coffea cenephora) in inhibiting the growth of Streptococcus mutans bacteria in vitro does not provide significant results through the dilution method, but the results show a decrease in the growth of the number of Streptococcus mutans colonies.
