Abstract
Latar belakang: Makrofag adalah sel fagosit mononuklear yang melepaskan IL-1β, sitokin proinflamasi yang dilepaskan sebagai respon pertahanan terhadap cedera. Gel ekstrak buah ciplukan mengandung senyawa aktif salah satunya yaitu lupeol, memiliki kemampuan antiinflamasi melalui penghambatan sintesis sitokin proinflamasi. Tujuan: Mengetahui perbedaan pemberian triamcinolone acetonide dengan pemberian gel ekstrak buah ciplukan (Physalis angulata L.) terhadap jumlah makrofag dan ekspresi seluler IL-1β pada penyembuhan luka ulkus traumatik mukosa labial tikus putih. Metode: Penelitian ini dibagi 9 kelompok dengan 3 time series (hari ke-2, -6 dan -12) dan dibagi 3 kelompok perlakuan (kontrol, ekstrak buah ciplukan, triamcinolone acetonide), semua tikus diinduksi ulkus traumatik. Dilakukan pembuatan sediaan dan pewarnaan HE dan IHK. Pengamatan sediaan menggunakan mikroskop digital. Penghitungan jumlah makrofag dan IL-1β menggunakan OLYVIA dan ImageJ. Hasil: Analisis data menggunakan uji independent t- test, menunjukkan perbedaan yang signifikan ekspresi seluler IL-1β antara kelompok physalis dan triamcinolone pada hari ke-2, ke-6 dan ke-12. Uji korelasi pearson menunjukkan semakin lama perlakuan pada 3 time series (hari ke-2, ke-6 dan ke-12) yang dilakukan dapat menurunkan jumlah makrofag dan IL-1β dan didapatkan hubungan jumlah makrofag dengan IL-1β pada semua kelompok. Kesimpulan: Terdapat perbedaan antara pemberian triamcinolone acetonide dengan pemberian gel ekstrak buah ciplukan (Physalis angulata L.) terhadap ekspresi seluler interleukin 1 beta (IL-1β) pada penyembuhan luka ulkus traumatik mukosa labial tikus putih (Rattus norvegicus).
English Abstract
Background: Macrophage is a phagocytic mononuclear cell that release IL-1β, a proinflammatory cytokine that is released as a host defense response to injury. Physalis angulata fruit extract gel contains active compounds which one is lupeol, has anti- inflammatory ability by inhibiting the synthesis of proinflammatory cytokines. Objective: To determine the comparison of the administration of Triamcinolone acetonide with Physalis angulata fruit extract gel to the number of macrophages and cellular expression of IL-1β in labial mucous traumatic ulcers wound healing of white rats. Method: This study was divided into 9 groups with 3 time series (days -2, -6 and -12), and divided into 3 treatment groups (control, physalis angulata fruit extract gel, triamcinolone acetonide), were induced by traumatic ulcers. The preparation and staining use HE and IHC. Observation of preparations using a digital microscope. Macrophage and IL-1β counts are calculated using OLYVIA and ImageJ. Results: Data analysis using independent t-test, showed significant differences in the cellular expression of IL-1β between physalis and triamcinolone groups on days 2, 6 and 12. Pearson correlation test showed that the longer treatment in 3 time series (2nd, 6th and 12th) that was carried out could reduce the number of macrophages and IL-1β and the relationship between the number of macrophages and IL-1β in all groups. Conclusion: There is a difference between the administration of triamcinolone acetonide with physalis angulata fruit extract gel to the cellular expression of interleukin 1 beta (IL-1β) in labial mucous traumatic ulcers wound healing of white rats (Rattus norvegicus).
