Abstract
Perak nitrat secara medis digunakan sebagai anti infeksi, antiseptik dan obat topikal. Perak nitrat (AgNO3) yang digunakan secara berlebihan dapat mengganggu kondisi fisiologis tubuh. Efek yang ditimbulkan dari sifat toksik perak nitrat yaitu dapat mengganggu kondisi ginjal. Ginjal berfungsi untuk mengatur metabolisme, mengontrol volume dan komposisi cairan tubuh. Fungsi ginjal dapat terganggu karena kerusakan komponennya. Gangguan fungsi ginjal dapat dicegah dengan pemberian antioksidan. Antioksidan dapat menghambat dan menetralisir terjadinya reaksi oksidasi yang melibatkan radikal–radikal bebas. Madu kelengkeng mengandung antioksidan seperti vitamin C, vitamin E dan flavonoid yang dapat mencegah kerusakan ginjal. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian madu kelengkeng pada mencit (Mus musculus) terhadap kadar BUN dan kreatinin yang diinduksi perak nitrat (AgNO3). Penelitian ini bersifat percobaan menggunkan Rancangan Acak Lengkap, dilakukan dengan menginduksi larutan AgNO3 secara peroral pada 20 ekor mencit jantan, dengan berat 25-30 gram, berumur 2-3 bulan selama 14 hari. Penelitian ini dibagi menjadi empat kelompok perlakuan yaitu P1(kontrol negatif, tidak dilakukan pemberian madu kelengkeng dan perak nitrat), P2 (kontrol positif, pemberian perak dosis 200 μg/0,5ml), P3 (placebo, pemberian madu kelengkeng dosis 0.08mL/20g) dan P4 (perlakuan pencegahan, pemberian perak dosis 200 μg/0,5ml dan madu kelengkeng dosis 0.08mL/20g) dengan masing-masing kelompok terdapat 5 pengulangan. Pengambilan data berupa kadar BUN dan Kreatinin dilakukan dengan menggunakan metode spektofotometri. Data tersebut kemudian dianalisis secara kuantitatif menggunakan uji One Way Analysis of Varlance (ANOVA) dan dilanjutkan dengan uji Tukey. Berdasarkan hasil penelitian yang dilakukan dapat disimpulkan pada perlakuan 4 (P4) pemberian madu kelengkeng (volume 0,4 mL dengan dosis 0,08mL/20g) dan perak nitrat dengan dosis 200 μg/0,5ml dapat mencegah peningkatan kadar BUN (P<0,05) dan belum dapat mencegah peningkatan kadar kreatinin pada mencit (Mus musculus) (P>0,05).
English Abstract
Silver nitrate is medicinally used as an anti-infective, antiseptic and topical medicine. Silver nitrate (AgNO3) which is used in excess can disrupt the physiological condition of the body. The effect of silver nitrate's toxic properties is that it can interfere with kidney conditions. The kidneys function to regulate metabolism, control the volume and composition of body fluids. Kidney function can be impaired due to damage to its components. Impaired kidney function can be prevented by giving antioxidants. Antioxidants can inhibit and neutralize oxidation reactions that involve free radicals. Longan honey contains antioxidants such as vitamin C, vitamin E and flavonoids which can prevent kidney damage. This study aims to determine the effect of longan honey on mice (Mus musculus) on levels of BUN and creatinine induced by silver nitrate (AgNO3). This research is an experimental study using a completely randomized design, carried out by inducing a solution of AgNO3 orally in 20 male mice, weighing 25-30 grams, 2-3 months old for 14 days. This study was divided into four treatment groups, namely P1 (negative control, no longan honey and silver nitrate), P2 (positive control, 200 μg / 0.5ml silver), P3 (placebo, 0.08mL / Longan honey). 20g) and P4 (preventive treatment, giving silver with a dose of 200 μg / 0.5ml and longan honey at a dose of 0.08mL / 20g) with 5 repetitions of each group. Data were collected in the form of BUN and creatinine levels using spectophotometric methods. The data were then analyzed quantitatively using the One Way Analysis of Varlance (ANOVA) test and continued with the Tukey test. Based on the results of the research conducted, it can be concluded that in treatment 4 (P4), giving longan honey (volume 0.4 mL at a dose of 0.08 mL / 20g) and silver nitrate at a dose of 200 μg / 0.5ml can prevent an increase in BUN levels (P <0 , 05) and has not been able to prevent the increase in creatinine levels in mice (Mus musculus) (P> 0.05).
