Abstract

Coronavirus disease 2019 (COVID-19) adalah penyakit yang disebabkan oleh infeksi severe acute respiratory syndrome corona virus 2 (SARS-CoV-2). Vaksinasi adalah salah satu tindakan pencegahan dan pengendalian penyebaran penyakit COVI D-19. Highly conserved region (HCR) dan receptor binding domain (RBD) merupakan bagian protein spike dalam virus SARS-CoV-2 yang berpotensi sebagai kandidat vaksin karena cenderung stabil tidak mutasi dan dapat dikenali oleh strain virus corona baru. Tujuan dari penelitian ini adalah untuk mengetahui bahwa imunisasi vaksin subunit spike SARS-CoV-2 berbasis Lactococcus lactis dapat menginduksi respon imun IgG anti-spike SARS-CoV2 pada mencit BALB/c. 49 ekor mencit BALB/c jantan usia 6-8 minggu dibagi menjadi 7 kelompok perlakuan, yaitu kontrol negatif (tikus tanpa perlakuan) dan 6 kelompok perlakuan.Tikus pada semua kelompok perlakuan diberikan pakan diet standar AIN-93M selama 56 hari dan diimunisasi Lactococcus lactis (NZ3900) yang mengandung plasmid pNZ8149-HCR dan pNZ8149-RBD selama tiga hari berturut-turut (hari ke1-3, 14-16, 28-30) melalui rute oral dan intranasal. Pada hari ke-57 mencit dikorbankan dan sampel darah diambil untuk diukur kadar IgG anti-spike SARS-CoV-2 serum menggunakan Uji ELISA. Analisis statistik menunjukkan bahwa pemberian vaksin L. lactis yang mengekspresikan protein spike SARS-CoV-2 dapat menginduksi imun IgG anti-spike SARS-CoV-2 secara signifikan (p< 0,05). Kesimpulan penelitian ini adalah bahwa pemberian vaksin subunit spike SARS-CoV-2 berbasis L. lactis dapat menginduksi respon imun IgG anti-spike SARS-CoV2 pada mencit BALB/c dan pemberian vaksin melalui rute nasal lebih tinggi menginduksi respon imun IgG anti-spike SARS-CoV-2 daripada rute oral.

English Abstract

Coronavirus disease 2019 (COVID-19) is caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Vaccination is one of the tools to prevent and control COVID-19. Highly conserved region (HCR) and receptor binding domain (RBD) are proteins in the SARS-CoV-2 virus that are potential as vaccine candidates because it tends to be stable, not mutated and can be recognized by new corona virus strains. This study aimed to analyze the Lactococcus lactis which expressing SARS-CoV-2 spike subunit vaccine immunization could induce IgG anti-spike SARS-CoV2 immune response in BALB/c mice. Forty nine male BALB/c mice aged 6-8 weeks were divided into 7 treatment groups, namely negative control (rats without treatment) and 6 treatment groups. Mice in all treatment groups were given standard diet AIN-93M for 56 days and immunized with Lactococcus lactis (NZ3900) containing the plasmids pNZ8149-HCR and pNZ8149-RBD for three consecutive days (days 1- 3, 14-16, 28-30) via the oral and intranasal routes. On day 57 the mice were sacrificed and blood samples were taken to measure the serum IgG anti-spike SARS-CoV-2 level using the ELISA test. Statistical analysis showed that administration of L. lactis vaccine expressing the SARS-CoV-2 spike protein could significantly induce SARS-CoV-2 anti-spike IgG immunity p<0.05). The study concludes that the administration of the SARS-CoV-2 spike subunit vaccine based on L. lactis can induce an IgG anti-spike SARS-CoV2 immune response in BALB/c mice and that administration of the intranasal vaccine induces a higher IgG agains-spike SARS-CoV-2 immune response than the oral route.
