Abstract

Anestesi adalah tindakan pengeliminasian rasa sakit dalam tubuh dari kegiatan yang mengakibatkan sakit. Agen anestesi yang biasanya digunakan ialah injeksi agen anestesi kombinasi ketamin-xylazin dan ketamin-diazepam. Tujuan penelitian ini ialah mengetahui pengaruh agen anestesi kombinasi ketamin-xylazin dan ketamin-diazepam terhadap morfologi eritrosit dan diferensial leukosit pada burung lovebird. Penelitian ini menggunakan 18 ekor burung lovebird yang terbagi menjadi dua kelompok, yaitu kelompok perlakuan 1 yang diberi anestesi ketamindiazepam dengan masing-masing dosis 30 mg/kgBB dan 2 mg/kgBB serta kelompok perlakuan 2 yang diberi anestesi ketamin-xylazin dengan masing-masing dosis 50 mg/kgBB dan 10 mg/kgBB. Pemberian volume anestesi dilakukan dengan cara injeksi pada musculus pectoralis. Pengambilan sampel darah dilakukan melalui vena brachialis lalu dilakukan ulas darah pada object glass. Pengamatan morfologi eritrosit dan diferensial leukosit dilakukan menggunakan mikroskop optilab perbesaran 1000x. Data morfologi eritrosit dijelaskan secara deskriptif sedangkan diferensial leukosit diolah dengan Wilcoxon Signed Rank Test. Terjadi perubahan morfologi berupa inti sel atau nukleus eritrosit yang memudar pada 22,2% sampel eritrosit dan 77,8% eritrosit tidak mengalami perubahan morfologi pada bentuk, warna, dan ukuran eritrosit. Semua data diferensial leukosit menunjukkan nilai probabilitas p >0,05. Baik ketamin-xylazin dan ketamindiazepam menunjukkan perubahan morfologi berupa pemudaran inti pada sebagian kecil eritrosit, tetapi tidak mempengaruhi diferensial leukosit lovebird.

English Abstract

Anesthesia is the act of eliminating pain in the body from activities that cause pain. The anesthetic agent that is usually used is the injection of a combination of ketamine-xylazine and ketamine-diazepam anesthetic agents. The purpose of this study was to determine the effect of a combination of ketaminexylazine and ketamine-diazepam anesthetic agents on erythrocyte morphology and leukocyte differential in lovebird. This study was conducted with 18 lovebirds which were divided into two groups, namely 1st treatment group which was given ketamine-diazepam anesthesia with each dose of 30 mg/kg and 2 mg/kg and 2nd treatment group which was given ketamine-xylazine anesthesia with each dose of 50 mg/kg and 10 mg/kg. The volume of anesthesia was given by injection into the pectoral muscle. Blood samples were taken through the brachial vein and then carried out a blood smear on the object glass. Observation of erythrocyte morphology and leukocyte differential was carried out using an optilab microscope with 1000x magnification. Erythrocyte morphological data were described descriptively while the leukocyte differential was processed by the Wilcoxon Signed Rank Test. Morphological changes occurred in the form of cell nuclei or erythrocyte nuclei that faded in 22.2% erythrocytes and 77.8% erythrocytes did not experience morphological changes in the shape, color, and size of erythrocytes. All leukocyte differential data showed p probability value > 0.05. Both ketaminexylazine and ketamine-diazepam showed morphological changes in the form of nuclear fading in a small number of erythrocytes, but did not affect the lovebird leukocyte differential.
