Abstract
Salmonella sp. merupakan bakteri patogen di saluran pencernaan, pada Infectious Dose (ID) >106 CFU mampu menimbulkan gejala klinis. Probiotik (Lactobacillus sp., Enterococcus sp., Streptococcus sp., Bacillus dan Saccharomyces cerevisiae) merupakan bakteri, fungi atau yeast yang mempunyai efek menguntungkan bagi hospes dengan meningkatkan mikroflora dalam saluran pencernaan. Manfaat probiotik adalah mempertahankan barier mukosa usus dan dapat mencegah adhesi Salmonella sp. pada mukosa usus. Tujuan penelitian ini untuk mengetahui perbedaan histomorfometri ketebalan mukosa dan kedalaman kripta jejunum mencit yang diberi probiotik dan ditantang Salmonella sp. dan pengaruh pemberian probiotik terhadap histomorfometri histopatologi jejunum mencit yang ditantang Salmonella sp. Penelitian ini menggunakan 20 ekor mencit yang dibagi dalam empat kelompok perlakuan: P1 (probiotik+Salmonella sp.), P2 (Probiotik), P3 (Salmonella sp.), P4 (tanpa perlakuan). Identifikasi BAL dianalisis secara deskriptif menggunakan media deMann Rogosa Sharpe Agar (MRSA), pewarnaan Gram, dan uji katalase. Histomorfometri jejunum meliputi pengukuran ketebalan mukosa dan kedalaman kripta dianalisis secara kuantitatif menggunakan uji One Way Anova yang dilanjutkan dengan uji Duncan. Skoring lesi dianalisa dengan statistik non-parametrik menggunakan Kruskall Wallis dan Mann-Whitney. Identifikasi koloni hasil penelitian adalah koloni bulat berwarna putih, morfologi bakteri berbentuk basil, bersifat Gram positif, katalase negatif bakteri asam laktat kelompok Lactobacillus spp., sementara pada pengamatan histopatologi, meliputi skoring lesi dan histomorfometri tebal mukosa dan kedalaman kripta terdapat perbedaan yang signifikan (p<0,05) pada masing-masing perlakuan. Kesimpulan dari penelitian ini yaitu terdapat perbedaan yang signifikan di antara rerata skoring lesi dan histomorfometri ketebalan mukosa serta kedalaman kripta masing-masing perlakuan, namun pada uji lanjutan diketahui bahwa perlakuan pemberian preventif probiotik tidak berbeda nyata (p<0,05) dengan kontrol. Sehingga pada penelitian ini diketahui bahwa pemberian probiotik sebagai preventif masih dapat menimbulkan kerusakan mukosa jejunum mencit (Mus musculus) yang ditantang Salmonella sp.
English Abstract
Salmonella sp. is a pathogenic bacteria in the digestive tract, the Infectious Dose (ID)> 106 CFU can cause clinical symptoms. Probiotics (Lactobacillus sp., Enterococcus sp., Streptococcus sp., Bacillus and Saccharomyces cerevisiae) are bacteria, fungi or yeast that have beneficial effects on the host by increasing the microflora in the digestive tract. The benefit of probiotics is to maintain the intestinal mucosal barrier and can prevent the adhesion of Salmonella sp. on the intestinal mucosa. The purpose of this study was to discover the histomorphometric differences in mucosal thickness and depth of jejunal crypt of mice given probiotics and challenged by Salmonella sp. and the effect of probiotics on the histopathological histomorphometry of the mice challenged by Salmonella sp. This study used 20 mice divided into four treatment groups consisting of: P1 (probiotics + Salmonella sp.), P2 (Probiotics), P3 (Salmonella sp.), P4 (without treatment). The identification of LAB was analyzed descriptively using deMann Rogosa Sharpe Agar (MRSA) media, Gram stain, and catalase test. Jejunum histomorphometry including measurement of mucosal thickness and depth of crypt was analyzed quantitatively using the One Way Anova test followed by Duncan's test. The lesion scoring was analyzed using non-parametric statistics using Kruskall Wallis and Mann-Whitney. The results of this study found white round colonies, bacilli-shaped, Gram positive, catalase negative lactic acid bacteria from the Lactobacillus spp. Group, while in histopathological observations through lesion scoring and histomorphometric of mucosal thickness and depth of crypt, there was a significant difference (p <0.05) in each treatment. The conclusion of this study is that there is a significant difference between the mean lesion scoring and histomorphometry of mucosal thickness and crypt depth of each treatment, but in the follow-up test it was found that the probiotic preventive treatment was not significantly different (p<0.05) with the control. So that in this study it was known that probiotics as a preventive can still cause damage to the jejunum mucosa of mice (Mus musculus) which were challenged by Salmonella sp.
