Abstract

Konsumsi makanan yang mengandung high-fat-high-fructose (HFHF) dapat menyebabkan obesitas. Obesitas dapat menyebabkan aktivasi kronis dari jalur inflamasi pada adiposit dan makrofag yang berada di dalam jaringan adiposa. Jaringan adiposa pada kondisi obesitas diperkaya dengan sitokin proinflamasi, yang mana salah satunya adalah TNF-alfa. Sitokin proinflamasi akan memicu respons inflamasi. Jamur Tiram (Pleurotus ostreatus) mengandung senyawa betaglukan dengan efek farmakologis yang luas, seperti antiobesitas, antiinflamasi, dan modulasi imun. Tujuan penelitian ini adalah untuk mengetahui pengaruh ekstrak beta-glukan jamur tiram terhadap kadar TNF-alfa serum pada tikus yang diinduksi diet HFHF. Hewan coba yang digunakan pada penelitian ini adalah 40 tikus Sprague dawley jantan yang dibagi ke dalam empat kelompok perlakuan yang berbeda, yaitu kelompok KN (diet normal AIN-93M), KP (diet HFHF modifikasi AIN-93M), P1 (diet HFHF modifikasi AIN-93M + larutan ekstrak betaglukan 125 mg/kgBB), dan P2 (diet HFHF modifikasi AIN-93M + larutan ekstrak beta-glukan 375 mg/kgBB). Beta-glukan yang didapatkan dari ekstraksi jamur tiram diidentifikasi gugus aktifnya menggunakan spektroskopi FTIR dan dianalisis bobot kandungannya menggunakan metode Megazyme. Pengukuran kadar TNFalfa serum dilakukan dengan metode ELISA. Hasil menunjukkan bahwa kadar TNF-alfa (ng/L ± SD) pada kelompok perlakuan yang diberi ekstrak beta-glukan lebih rendah (P1 = 83,00 ± 3,47, P2 = 84,21 ± 6,80) daripada kelompok perlakuan yang lain (KN = 91,52 ± 6,61, KP = 88,58 ± 5,01). Hasil analisis data dengan uji korelasi Spearman untuk mengetahui korelasi antar variabel menunjukkan bahwa ada korelasi yang bermakna antara beta-glukan dengan kadar TNF-alfa serum (p = 0,006, r = -0,445).

English Abstract

Consumption of foods containing high-fat-high-fructose (HFHF) can lead to obesity. Obesity can cause chronic activation of the inflammatory pathways in both adipocytes and macrophages located in adipose tissue. Adipose tissue in obesity is enriched with proinflammatory cytokines, which one of them called TNF-alpha. Proinflammatory cytokines trigger the inflammatory response. Oyster mushroom (Pleurotus ostreatus) contains beta-glucan compound with a broad pharmacological effects, such as antiobesity, antiinflammatory, and immunomodulatory. The purpose of this study was to determine the effects of oyster mushroom extract containing beta-glucan on rats induced by the HFHF diet. The animals used in this study were 40 male Sprague dawley rats which were assigned into four different treatment groups, categorized as KN (AIN-93M normal diet), KP (AIN-93M-modified HFHF diet), P1 (AIN-93M-modified HFHF diet + 125 mg/kgBW of beta-glucan extract solution), and P2 (AIN-93M-modified HFHF diet + 375 mg/kgBW of beta-glucan extract solution). Beta-glucan that obtained from the extraction of oyster mushrooms was identified its active groups using FTIR spectroscopy and its content weight was analyzed by the Megazyme method. Measurement of serum TNF-alpha levels was carried out using ELISA method. The results showed that TNF-alpha levels (ng/L ± SD) in the treatment groups that were given beta-glucan extract (P1 = 82.99 ± 3.47, P2 = 84.21 ± 6.80) were lower than the other treatment groups (KN = 91.52 ± 6.61, KP = 88.58 ± 5.01). The results of data analysis using Spearman’s correlation to find out the correlation between variables indicated that there was a significant correlation between beta-glucan and serum TNF-alpha levels (p = 0.006, r = -0.445).
