Abstract

Inflammatory Bowel Disease (IBD). IBD merupakan kondisi idiopatik kronis dari inflamasi intestinal yang menyebabkan gejala diare, nyeri abdomen, pendarahan, anemia, dan penurunan berat badan. Salah satu penyebab dari IBD meliputi pemberian Non-steroid Anti Inflammatory Drugs (NSAID) berlebihan dalam jangka waktu panjang. Anthocyanin merupakan bahan aktif utama yang terkandung dalam blueberry. Anthocyanin memiliki efek anti-inflamasi, antioksidan, dan antimikroba. Tujuan dari penelitian ini ialah untuk mengetahui pengaruh preventif dari ekstrak blueberry terhadap jumlah sel radang duodenum serta jumlah neutrofil darah tikus putih (Rattus norvegicus) model Inflammatory Bowel Disease (IBD) yang diinduksi Indometasin. Penelitian ini dilakukan kepada 5 kelompok tikus Wistar yang masing-masing kelompok terdiri atas 5 ekor tikus. Kelompok tersebut terbagi menjadi kelompok kontrol negatif, kontrol positif yaitu diberikan Indometasin 15 mg/kg BB, kelompok P1 yaitu diberikan ekstrak blueberry 1000 mg/kgBB dan Indometasin, P2 diberikan ekstrak blueberry 1500 mg/kgBB dan Indometasin, serta P3 diberikan ekstrak blueberry 2000 mg/kgBB dan Indometasin. Analisis data penelitian ini menggunakan analisis kuantitatif statistik ANOVA (Analysis of Variance) satu arah dengan tingkat kepercayaan 95% (p<0,05) dan dilanjutkan dengan uji Tukey untuk mengetahui hasil yang paling berbeda nyata. Hasil uji ANOVA jumlah sel radang menunjukkan perbedaan yang signifikan di antara kelompok perlakuan, dimana kelompok P1 (1500 mg/kgBB) berbeda signifikan terhadap K-, dan K+ berbeda signifikan dengan K- dan P3 (2000 mg/kgBB). Hasil uji ANOVA jumlah neutrofil menunjukkan tidak terdapat perbedaan yang signifikan di antara kelompok perlakuan. Hal ini diduga disebabkan karena terdapat faktor stress kronis yang dikonfirmasi dengan adanya peningkatan pada rasio neutrofil limfosit (NLR). Kesimpulan penelitian ini adalah terapi ekstrak blueberry kurang mampu mencegah peningkatan nilai neutrofil darah, namun mampu mencegah peningkatan jumlah sel radang pada histopatologi duodenum tikus putih (Rattus norvegicus), dengan dosis yang paling baik pada P3 yaitu 2000 mg/kg BB.

English Abstract

Inflammatory Bowel Disease (IBD) is a chronic idiopathic disease that cause significant clinical signs including diarrhea, abdominal pain, intestinal hemorrhage, anemia, and weight loss. One of the main causes in IBD is excessive use of Non-inflammatory Bowel Disease (NSAID). Anthocyanin is a main active compound that contains in blueberry. Anthocyanin have anti inflammatory, antioxidant, antimicrobe effect, as well as inhibiting chemotaxis movement including neutrophil. The main purpose of this research is to acknowledge the preventive benefit of blueberry extract according to inflammatory cell count of duodenum and neutrophil level of White Rats (Rattus Norvegicus) IBD model induced by Indomethacin. In this research, white rats (Rattus norvegicus) 5 groups of white rats were used, which each group contains 5. K- group was a group of negative control rats, K+ group was a group of positive control rats that was given Indomethacin 15 mg/kg BW, P1 group was given 1000 mg/kg BW blueberry extract and induced with 15 mg/kg BW Indomethacin, P2 group was given 1500 mg/kg BW blueberry extract and induced with 15 mg/kg BW Indomethacin, and P3 group was given 2000 mg/kg BW blueberry extract and induced with 15 mg/kg BW Indomethacin. This research’s data was analysed by ANOVA (Analysis of Variance) quantitative statistical analysis with confidence level 95% (P<0,05) then continued with Tukey test to determine the most significant results among all. The results of the ANOVA test for the inflammatory cell count showed that there were several significant difference between the treatment group, which P1 group (1000 mg/kg BW) was significantly different from K- group, and K+ was significantly different from K- group and P3 group. The results of the ANOVA test for the neutrophil level showed that there were no significant difference between the treatment group. This result was assumed due to the internal factor of which then confirmed with elevated level of neutrophil-lymphocyte ratio (NLR). The conclusion of this research was that the blueberry extract as a therapy for IBD was less effective if assessed from neutrophil level, but effective if assessed from inflammatory cell count on histopathology of duodenum, according to ANOVA test with the best dose of 2000 mg/kg BW.
