Abstract

Klebsiella pneumonia ESBL menjadi ancaman keselamatan manusia akibat angka resistensi antibiotik yang terus meningkat. Klebsiella pneumoniae ESBL juga merupakan salah satu patogen penyebab infeksi pada seting pelayanan kesehatan. Peningkatan angka resistensi yang tidak seimbang dengan pengembangan antibiotik menimbulkan kekhawatiran. Dibutuhkan suatu inovasi agen antimikroba yang mudah didapatkan serta melimpah. Tanaman Serai (Cymbopogon citratus) merupakan rumput abadi yang tersebar luas di wilayah tropis dan subtropis dunia. Tanaman Serai sudah lama dimanfaatkan menjadi tanaman herbal selain kegunaan utamanya sebagai bumbu masakan. Penelitian ini menggunakan penelitian eksperimental laboratorium dengan desain true experimental. Fokus penelitian ditujukan pada keadaan Klebsiella pneumoniae ESBL setelah pemberian ekstrak etanol Tanaman Serai (Cymbopogon citratus) secara in vitro melalui metode dilusi agar untuk menentukan pertumbuhan koloni pada tiap plate. Kelompok perlakuan pada penelitian ini dibagi menjadi 2 kelompok yaitu kelompok kultur K. Pneumoniae ESBL dengan aquades (kontrol negatif) serta kelompok kultur K. Pneumoniae ESBL dengan Tanaman Serai 2.5%, 5%, 7.5%, 10%, 12.5%, 15%, 17.5%, 20%, 22.5%, 25%, 27.5%, dan 30%. Hasil penelitian menunjukkan konsentrasi 2.5% hingga 15% tidak didapatkan penurunan pertumbuhan koloni. Konsentrasi 15%< dimasukkan kedalam analisa statistik. Analisa statistik didapatkan nilai uji Kruskal Wallis sebesar 0.00 (p<0.05). Analisis data dilanjutkan menggunakan Uji Mann Whitney. Hasil analisis menunjukkan bahwa konsentrasi ekstrak Tanaman Serai (Cymbopogon citratus) 0% atau kontrol negatif menghasilkan hasil yang berbeda signifikan terhadap konsentrasi 22.5%, 25%, 27.5%, dan 30% namun tidak signifikan terhadap konsentrasi 17.5% dan 20%. Perbandingan antarkelompok lainnya menunjukkan nilai signifikansi. Uji signifikansi Sparman menghasilkan angka 0.000 dengan koefisien korelasi -0.982. Hasil tersebut menunjukkan peningkatan konsentrasi ekstrak Tanaman Serai akan menurunkan pertumbuhan koloni Klebsiella pneumoniae ESBL secara signifikan. Sehingga, dapat disimpulkan bahwa ekstrak Tanaman Serai (Cymbopogon citratus) memiliki efek antimikroba terhadap bakteri Klebsiella pneumonia ESBL secara in vitro

English Abstract

Klebsiella pneumonia ESBL is a threat to human safety due to the increasing number of antibiotic resistence. Klebsiella pneumoniae ESBL is also a pathogen causing infection in health care settings. The unbalanced increase in resistance rates with antibiotic development is cause for the concern recently. An innovative antimicrobial agent that is easily available and abundant is needed. Plant lemongrass (Cymbopogon citratus) is a perennial grass that is widespread in tropical and subtropical regions world. Lemongrass plant has long been used as a herbal plant besides its main use as a cooking spice. This research uses experimental laboratory research with true experimental design. The focus of the research was on the condition of Klebsiella pneumoniae ESBL after giving the ethanol extract of lemongrass (Cymbopogon citratus) in vitro through the agar dilution method to determine the growth of colonies on each plate. The treatment group in this study was divided into 2 group: culture group of K. pneumoniae ESBL with distilled water (negative control) and culture group K. Pneumoniae ESBL with 2.5 %, 5%, 7 .5 %, 10 %, 12.5 %, 1 5 %, 17.5%, 20%, 22.5%, 25%, 27.5% and 30% of lemongrass plant extract. The results showed a concentration of 2.5% to 15% did not show a decrease in colony growth. The concentration <15% was entered into the statistical analysis. Statistical analysis showed that the Kruskal Wallis test value was 0.00 (p <0.05). Data analysis was continued using the Mann Whitney test. The results of the analysis showed that the concentration of citronella plant extract (Cymbopogon citratus) 0% or negative control resulted in significantly different results at concentrations of 22.5%, 25%, 27.5%, and 30% but not significant at concentrations of 17.5% and 20%. The comparison between other groups shows a significance value. The Spearman significance test yields the number 0.000 with a correlation coefficient of -0.982. These results indicate that increasing the concentration of the extract of the Lemongrass plant will significantly reduce the growth of the Klebsiella pneumoniae ESBL colonies. Thus, it can be concluded that the extract of the Citronella Plant (Cymbopogon citratus) has an in vitro antimicrobial effect on the Klebsiella pneumonia ESB
