Abstract

Kombinasi fitobiotik jahe merah (Zingiber officinale var. Rubrum), kunyit (Curcuma longa Linn.), dan temulawak (Curcuma xanthorrhiza Roxb.) berkhasiat sebagai antiinflamasi. TLR2 merupakan reseptor yang terlibat dalam proses inflamasi. IL-6 merupakan mediator inflamasi yang dihasilkan melalui jalur pensinyalan TLR2. Penelitian ini bertujuan untuk melihat interaksi enam senyawa yaitu gingerol-6, paradol-6, kurkumin, bisdemetoksikurkumin, demetoksikurkumin, dan tetrahidrokurkumin terhadap reseptor TLR2 Gallus gallus domesticus menggunakan uji in silico berupa penambatan molekuler serta membuktikan bentuk ikatan penghambatan pada reseptor melalui respon biologis berupa penurunan kadar IL-6 pada serum darah menggunakan uji ELISA. Uji in vivo dilakukan menggunakan Rancangan Acak Lengkap (RAL) terhadap 20 ekor ayam broiler strain cobb slow feather dengan berat rata-rata awal 51,5 gram. Ayam dibagi dalam 5 perlakuan dengan penambahan kombinasi serbuk jahe merah, kunyit, dan temulawak (P1: 0,5%; P2: 1%; P3: 1,5%; P4: 2%; K(-): 0%). Hasil eksplorasi enam senyawa aktif secara in silico menunjukkan bahwa keenam senyawa yang dijadikan ligan mampu berinteraksi dengan reseptor TLR2 Gallus gallus domesticus. Kurkumin memiliki nilai energi ikatan paling rendah ketika berikatan dengan reseptor TLR2 Gallus gallus domesticus dibandingkan dengan kelima ligan uji lain dalam uji in silico dengan skor energi ikatan (ΔG) yaitu -8,0 kkal/mol serta memiliki satu ikatan hidrogen dalam residu asam amino Thr291 pada situs aktif reseptor. Energi ikatan paling rendah dari kompleks ikatan kurkumin dan TLR2 Gallus gallus domesticus membentuk kestabilan energi yang paling baik saat berinteraksi. Hasil uji statistika terhadap kadar IL-6 menggunakan ONE WAY ANOVA dengan tingkat kepercayaan 95% (p<0,05) dan uji lanjutan berupa Tukey test menunjukkan bahwa penambahan kombinasi serbuk jahe merah, kunyit, dan temulawak mampu menurunkan kadar IL-6 secara signifikan pada dosis 1,5% dan 2%. Kesimpulan dari penelitian ini yaitu kombinasi serbuk jahe merah, kunyit, dan temulawak mampu berfungsi sebagai antiinflamasi dilihat dari uji in silico dan in vivo serta dosis optimal penambahan kombinasi serbuk jahe merah, kunyit, dan temulawak sebagai antiinflamasi adalah 1,5%

English Abstract

Phytobiotic combination of red ginger (Zingiber officinale var. Rubrum), turmeric (Curcuma longa Linn.), and javanese turmeric (Curcuma xanthorrhiza Roxb.) are efficacious as anti-inflammatory. TLR2 is a receptor involved in inflammatory processes. IL-6 is an inflammatory mediator produced through the signaling pathway TLR2. This study aims to look at the interaction of six compounds namely gingerol-6, paradol-6, curcumin, bisdemethoxycurcumin, demethoxycurcumin, and tetrahydrocurcumin to TLR2 Gallus gallus domesticus receptor using in silico test in the form of molecular docking and prove the form of inhibitory binding in receptor through biological response in the form of decreased IL-6 levels in blood serum using ELISA test. The in vivo test was conducted using a Complete Randomized Design (RAL) against 20 broiler chickens strain of cobb slow feather with an initial average weight of 51.5 grams. Chickens are divided into 5 treatments with the addition of a combination of red ginger powder, turmeric, and temulawak (P1: 0.5%; P2: 1%; P3: 1.5%; P4: 2%; K(-): 0%). The results of the exploration on six active compounds with in silico test showed that the six compounds used as ligands are able to interact with the TLR2 Gallus gallus domesticus receptor. Curcumin has the lowest binding energy value when bonded to the TLR2 Gallus gallus domesticus receptor compared to the other five test ligands in the in silico test with a bond energy score (ΔG) of -8.0 kcal/mol and has one hydrogen bond that forms the amino acid residue Thr291 in the receptor's active site. The lowest bond energy from the curcumin and TLR2 Gallus gallus domesticus complex make the best energy stability when they interact. The results of statistical tests on IL-6 levels using ONE WAY ANOVA with a confidence level of 95% (p<0.05) and follow-up tests in the form of Tukey test showed that the addition of a combination of red ginger, turmeric, and javanese turmeric was able to lower IL-6 levels significantly at doses of 1.5% and 2%. The conclusion of this study is the combination of red ginger, turmeric, and javanese turmeric powder is able to serve as an anti-inflammatory seen from the in silico and in vivo test. The optimal dose of adding the combination of red ginger, turmeric, and temulawak powder as an anti-inflammatory is 1.5%.
