Abstract

Sektor perikanan budidaya memiliki potensi yang sangat luas untuk dikembangkan terutama dalam memenuhi permintaan pasar yang semakin meningkat. Akan tetapi usaha ini sering mengalami kendala akibat serangan penyakit, salah satu contohnya yaitu bakteri Pseudomonas fluorescens yang dapat mengakibatkan penyakit Hemorrhagic septicemia. Pengobatan antibiotik yang sering dilakukan namun memberikan efek samping yaitu bakteri resisten. Upaya alternatif lain yang dapat dilakukan yaitu dengan penggunaan bahan ala-mi salah satu contohnya yaitu daun nangka (A. heterophyllus). Hal ini dikare-nakan kandungan daun nangka sebagai antibakteri alami. Penelitian ini bertujuan untuk mengetahui pengaruh dan dosis optimal pemberian ekstrak daun nangka (A. heterophyllus) terhadap pertumbuhan bak-teri P. fluorescens. Penelitian ini dilakukan di Laboratorium Sentra Ilmu Hayati (LSIH), Universitas Brawijaya, Malang pada bulan Maret-April 2021. Metode penelitian yang digunakan yaitu metode eksperimen untuk mengetahui secara langsung pengaruh pemberian berbagai dosis ekstrak daun nangka (A. heterophyllus) terhadap pertumbuhan bakteri P. fluorescens. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perla-kuan dan 3 ulangan. Dosis perlakuan ekstrak kasar daun nangka (A. heterophyl-lus) yaitu: A (60 ppm), B (120 ppm), C (180 ppm), D (240 ppm) dan E (300 ppm). Hasil penelitian menunjukkan bahwa ekstrak kasar daun nangka (A. heterophyl-lus) memberikan pengaruh sangat nyata terhadap daya hambat pertumbuhan bakteri P. fluorescens. Pengamatan zona hambat dilakukan setelah inkubasi selama 24 dan 48 jam. Hasil pengamatan setelah inkubasi selama 24 jam menunjukkan dosis ekstrak dengan rata-rata diameter zona bening tertinggi yaitu pada perlakuan C sebesar 11,48 mm dan dosis ekstrak dengan rata-rata zona bening terendah pada perlakuan E sebesar 8,85 mm. Pengamatan setelah inku-basi selama 48 jam didapatkan hasil bahwa zona bening mengecil yang menan-dakan sifat antibakteri sebagai bakteriostatik. Penelitian ini menunjukkan bahwa hubungan antara pemberian ekstrak kasar daun nangka (A. heterophyllus) ter-hadap pertumbuhan bakteri P. fluorescens membentuk pola kuadratik dengan persamaan y = -0,0001x2 + 0,05x + 6,327 dan koefisien nilai determinasi (R2) sebesar 0,70. Kesimpulan pada penelitian ini yaitu pemberian ekstrak kasar daun nang-ka (A. heterophyllus) efektif sebagai bahan antibakteri alami bakteri P. fluo-rescens. Bahan alami ini bersifat bakteriostatik, yaitu mampu menghambat per-tumbuhan bakteri. Perlakuan dosis yang optimal dari persamaan grafik hasil uji polynomial orthogonal yaitu pada dosis 250 ppm, sedangkan perlakuan dengan dosis 180 ppm pada uji cakram dengan hasil rerata zona hambat yaitu 11,48 mm menghasilkan nilai zona hambat tertinggi.

English Abstract

The cultivation sector has a very wide potential to be developed, especial-ly in meeting the increasing market demand. However, this effort is often ham-pered by disease attacks, one example is the bacterium Pseudomonas fluo-rescens which can cause Hemorrhagic septicemia. Antibiotic treatment is often done but has a side effect, namely bacterial resistance. Another alternative effort that can be done is by using natural ingredients, one example is jackfruit (A. het-erophyllus) leaves. This is because the content of jackfruit leaves as a natural antibacterial. This study aims to determine the effect and optimal dose of jackfruit leaf extract (A. heterophyllus) on the growth of P. fluorescens bacteria. This research was conducted at the Laboratorium Sentra Ilmu Hayati (LSIH), Universitas Brawi-jaya, Malang in March-April 2021. The research method used is an experimental method to determine di-rectly the effect of giving various doses of jackfruit leaf extract (A. heterophyllus) on the growth of P. fluorescens bacteria. This study used a completely random-ized design (CRD) with 5 treatments and 3 replications. The treatment doses of jackfruit leaf crude extract (A. heterophyl-lus) were: A (60 ppm), B (120 ppm), C (180 ppm), D (240 ppm) and E (300 ppm). The results showed that the crude ex-tract of jackfruit (A. heterophyllus) leaves had a very significant effect on the inhi-bition of the growth of P. fluorescens bacteria. The zone of inhibition was carried out after 24 and 48 hours of incubation. The results of observations after incuba-tion for 24 hours showed that the extract dose with the highest average clear zone diameter was 11.48 mm in treatment C and the extract dose with the lowest average clear zone diameter was 8.85 mm in treatment E. After incubation for 48 hours, the results showed that the clear zone was reduced which indicated its antibacterial properties as bacteriostatic. This study showed that the relationship between the crude extract of jackfruit leaves (A. heterophyllus) and the growth of P. fluorescens bacteria formed a quadratic pattern with the equation y = -0.0001x2 + 0.05x + 6.327 and the coefficient of determination (R2) of 0.70. The conclusion of this study is that the crude extract of jackfruit leaves (A. heterophyllus) is effective as a natural antibacterial agent for P. fluorescens bac-teria. This natural material is bacteriostatic, which is able to inhibit the growth of bacteria. The optimal dose treatment from the equation of the polynomial orthog-onal test results is at a dose of 250 ppm, while the treatment with a dose of 180 ppm in the disc test with an average inhibition zone of 11.48 mm produces the highest inhibition zone value.
