Abstract

Kanker merupakan salah satu penyakit yang paling ditakuti, karena proses pengobatan yang sangat mahal. Penyakit ini berkembang dengan cepat dan tidak terkendali melalui sel-sel pada jaringan tubuh yang tidak normal. Faktor dari penyakit kanker yaitu radiasi, virus, infeksi, rokok dan bahan kimia dari makanan. Salah satu pengobatan alternatif yang digunakan dalam penyembuhan penyakit kanker adalah menggunakan tumbuhan yang terdapat dilingkungan sekitar. Tumbuhan yang banyak digunakan dalam terapi penyembuhan penyakit kanker adalah mangrove. Pada penelitian ini menggunakan mangrove dengan spesies Avicennia marina. Kandungan yang dimiliki oleh tumbuhan mangrove antara lain senyawa bioaktif meliputi alkaloid, tanin, flavonoid, saponin dan terpenoid, terbukti memiliki aktivitas antikanker. Senyawa-senyawa yang terkandung mampu menghambat pertumbuhan sel kanker. Tujuan dari penelitian ini adalah untuk mengetahui nilai IC50 yang berasal dari ekstrak daun mangrove Avicennia marina. Metode yang digunakan dalam penelitian ini adalah metode eksprimen. Parameter uji meliputi rendemen, uji kadar air, uji fitokimia, uji toksisitas, uji total fenol, uji sitotoksisitas dan uji LC-MS. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) sederhana dan dilanjutkan dengan uji Tukey. Hasil penelitian didapatkan hasil rendemen pada ekstrak n-heksan sebesar 3,96%, ekstrak etil asetat sebesar 6,40% dan ekstrak etanol sebesar 14,40%. Hasil uji kadar air pada ekstrak n-heksan sebesar 11,99%, ekstrak etil asetat sebesar 12,80% dan ekstrak etanol sebesar 16,57%. Hasil uji fitokimia menunjukkan bahwa pada ekstrak n-heksan positif mengandung alkaloid, steroid/triterpenoid dan flavonoid. Pada ekstrak etil asetat positif mengandung steroid/triterpenoid dan tanin. Pada ekstrak etanol positif mengandung saponin, steroid/triterpenoid, flavonoid dan tanin. Uji toksisitas dengan metode BSLT diperoleh nilai hasil LC50 sebesar 98,55 ppm pada ekstrak etanol, 441,39 ppm pada ekstrak n-heksan dan 1931,49 ppm pada elstrak etil asetat. Pada hasil uji total fenol diperoleh nilai sebesar 113,22 mg/g GAE pada ekstrak n-heksan, 752,97 mg/g GAE pada ekstrak etil asetat dan 1915,92 mg/g GAE pada ekstrak etanol. Pengujian sitotoksisitas pada sel HeLa menggunakan metode MTT assay menghasilkan nilai viabilitas tertinggi pada dosis 62,5 ppm sebesar 56,800% dan hasil terendah pada dosis 1000 ppm sebesar 29,233%. Untuk dosis 500 ppm menghasilkan nilai viabilitas sebesar 36,233%, dosis 250 ppm sebesar 42,733% dan dosis 125 ppm sebesar 47,800%. Nilai IC50 yang dihasilkan dari penelitian ini sebesar 115,345 ppm yang menunjukan bahwa ekstrak etanol daun mangrove Avicennia marina cukup aktif terhadap sel HeLa. Hasil pengujian LC-MS menunjukkan dugaan senyawa Caffeine dan Diosmetin.

English Abstract

Cancer is one of the most feared diseases, because the treatment process is very expensive. This disease develops rapidly and uncontrollably through cells in body tissues that are not normal. Factors from cancer are radiation, viruses, infections, cigarettes and chemicals from food. One of the alternative treatments used to cure cancer is to use plants in the environment. Plants that are widely used in cancer healing therapy are mangroves. In this study using mangroves with Avicennia marina species. The content of mangrove plants includes bioactive compounds including alkaloids, tannins, flavonoids, saponins and terpenoids, proven to have anticancer activity. The purpose of this study was to determine the IC50 value derived from Avicennia marina leaf extract. The method used in this research is the experimental method. The test parameters include yield, water content test, phytochemical test, toxicity test, total phenol test, cytotoxicity test and LC-MS test. The experimental design used was a simple completely randomized design (CRD) and was followed by the Tukey test. The results showed that the yield of n-hexane extract was 3.96%, ethyl acetate extract was 6.40% and ethanol extract was 14.40%. The results of the water content test in the n-hexane extract were 11.99%, the ethyl acetate extract was 12.80% and the ethanol extract was 16.57%. The results of the phytochemical test showed that the positive n-hexane extract contained alkaloids, steroids / triterpenoids and flavonoids. The positive ethyl acetate extract contains steroids / triterpenoids and tannins. The positive ethanol extract contains saponins, steroids / triterpenoids, flavonoids and tannins. Toxicity test using the BSLT method obtained LC50 yield values of 98.55 ppm in ethanol extract, 441.39 ppm in n-hexane extract and 1931.49 ppm in ethyl acetate extract. In the total phenol test results obtained a value of 113.22 mg / g GAE in the n-hexane extract, 752, 97 mg / g GAE in ethyl acetate extract and 1915.92 mg / g GAE in ethanol extract.
