Abstract

Keberagaman aktivitas biologi yang dihasilkan oleh senyawa tiourea dan meningkatnya angka resistensi E. coli terhadap antibiotik mendorong dilakukannya pengembangan senyawa turunan tiourea sebagai agen antibakteri baru. Penelitian ini dilakukan dengan tujuan untuk mendapatkan senyawa 1-alil- 3-(4-klorobenzoil)tiourea yang memiliki aktivitas antibakteri terhadap E. coli dan membandingkan Konsentrasi Hambat Minimum (KHM) senyawa hasil sintesis dengan Konsentrasi Hambat Minimum (KHM) gentamisin terhadap E. coli. Sintesis senyawa dilakukan dengan metode Schotten-Baumann melalui reaksi substitusi nukleofilik, sedangkan uji aktivitas antibakteri dilakukan dengan metode dilusi agar. Hasil dari penelitian ini menunjukkan bahwa senyawa 1-alil-3-(4- klorobenzoil)tiourea dapat diperoleh dengan sintesis melalui metode Schotten- Baumann dan reaksi substitusi nukleofilik yang dikonfirmasi dengan uji konfirmasi struktur menggunakan Spektrofotometri Ultraviolet (UV), Spektrofotometri FTIR, Spektrometri NMR, dan Spektrometri Massa serta uji kemurnian senyawa menggunakan Kromatografi Lapis Tipis (KLT) dan Uji Rentang Titik Lebur. Hasil uji aktivitas antibakteri menunjukkan bahwa Konsentrasi Hambat Minimum (KHM) senyawa 1-alil-3-(4-klorobenzoil)tiourea tidak dapat ditentukan karena terdapat pertumbuhan bakteri pada seluruh seri konsentrasi, sedangkan gentamisin menunjukkan efek hambatan pada konsentrasi terkecil yakni 2 ppm. Hasil tersebut menyebabkan tidak dapat dilakukannya pembandingan Konsentrasi Hambat Minimum antara 1-alil-3-(4-klorobenzoil)tiourea dengan gentamisin. Dapat disimpulkan bahwa senyawa 1-alil-3-(4-klorobenzoil)tiourea dapat diperoleh melalui reaksi substitusi nukleofilik, namun tidak memiliki aktivitas antibakteri terhadap E. coli.

English Abstract

The diversity of biological activities produced by thiourea compounds and the increasing number of E. coli resistance to antibiotics encourage the development of thiourea derivatives as new antibacterial agents. The research was aimed to obtain 1-allyl-3-(4-chlorobenzoyl)thiourea compound which has antibacterial activity against E. coli and to compare the Minimum Inhibitory Concentration (MIC) of the synthesized compound with the Minimum Inhibitory Concentration (MIC) of gentamicin against E. coli. The compound synthesis was carried out by the Schotten-Baumann method through nucleophilic substitution reactions, while the antibacterial activity test was carried out by the agar dilution method. The results of this study showed that 1-allyl-3-(4-chlorobenzoyl)thiourea could be obtained by synthesis using Schotten-Baumann method and nucleophilic substitution reactions, confirmed by structural confirmation tests through Ultraviolet (UV) Spectrophotometry, Infrared Spectrophotometry (IR), NMR Spectrometry, and Mass Spectrometry, along with compound purity tests conducted by Thin Layer Chromatography (TLC) and Melting Point Range Test. The results of the antibacterial activity test showed that the Minimum Inhibitory Concentration (MIC) of the 1-allyl-3-(4-chlorobenzoyl)thiourea compound could not be determined because there was bacterial growth in the entire concentration series, while gentamicin showed an inhibitory effect at the smallest concentration of 2 ppm. These results made it impossible to compare the Minimum Inhibitory Concentrations (MIC) between 1-allyl-3-(4-chlorobenzoyl)thiourea and gentamicin. It can be concluded that 1-allyl-3-(4-chlorobenzoyl)thiourea can be obtained through a nucleophilic substitution reaction, but has no antibacterial activity against E. coli.
