Abstract
Fibrosis ginjal adalah tanda akhir dari kegagalan ginjal kronis. Ekstrak blueberry memiliki kandungan antosianin yang berperan sebagai antihipertensi dan antiinflamasi. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak blueberry terhadap ekspresi Interleukin - 6(IL-6) dan Glomerulosklerosis pada hewan model fibrosis ginjal induksi Unilateral Ureteral Obstruction (UUO). Mencit jantan strain Swiss webster dengan berat badan 20-30 gram, usia 6-8 minggu dibagi menjadi 5 kelompok perlakuan yaitu: Kontrol negatif tanpa perlakuan (Negatif), kontrol positif 7 hari (U7) dan 14 hari (U14) dengan penerapan prosedur UUO, dan perlakuan 7 hari (UB7) dan 14 hari (UB14) dengan penerapan prosedur UUO berterapi antosianin berdosis 1500mg/kgBB pemberian 1x24 jam. Pada akhir penelitian organ ginjal dikoleksi untuk pemeriksaan ekspresi interleukin - 6 (IL-6) dengan immunohistokimia dan skoring glomerulosklerosis. Data kuantiatif ekspresi IL-6 dianalisis secara menggunakan uji analisis ragam One Way ANOVA dengan lanjutan uji tukey atau uji Beda Nyata Jujur (BNJ). Kejadian glomerulosklerosis dinilai dengan sistem skoring dan dianalisis secara statistic non-parametrik Kruskall Wallis dan uji lanjutan Mann-Whitney.Hasil penelitian menunjukkan pemberian terapi ekstrak blueberry dosis 1500 mg/kg/BB/hari selama 7 dan 14 hari berpengaruh signifikan terhadap ekspresi IL-6 (p<0.05) dan skor glomerulosklerosis (p<0.05). Kesimpulan dari penelitian ini yaitu pemberian terapi ekstrak blueberry dosis 1500 mg/kg/BB/hari selama 7 hari dan 14 hari dapat menjaga rendahnya ekspresi IL-6 dan skor glomerulosklerosis ginjal.
English Abstract
Renal fibrosis is often described as an end stage of kidney failure. Blueberry extract known as anthocyanins act as antihypertensive and antiinflammatory properties. This research is aimed to acknowledge the effects of blueberry extract against Interleukin-6 (IL-6) expression and occurance of glomerular sclerosis in Unilateral Ureteral Obstruction (UU) induced fibrosis animal models. Mice of Swiss webster strain with 20-30 grams of body wegiht and 6-8 weeks of age are seperated into groups of 5: negative control group (Negative), positive control group of 7 (U7) and 14 days (U14) that are applied UUO procedure, and blueberry extract group of 7 (UB7) and 14 days (UB14) that are applied UUO procedure and therapied 1500 mg/kgBW blueberry extract daily. Kidney organ was collected for examination of interleukin-6 (IL-6) expression with immunohistochemsitry method and scoring of glomerular sclerosis. Quantitative data of IL-6 expression was analyzed initially with ANOVA test and continued with Tukey test. Assesment of glomerular Scelrosis occurance used a scoring system hereinafter analyzed with Kruskall Wallis' non-parametric statistics test and continued with Mann-Whitney test (alpha=0.05) . Research outcome showed blueberry extract dose 1500 mg/kg BW daily therapy of 7 and 14 days effects significantly (p<).05) againts IL-6 expression and glomerular sclerosis occurance. In conclusion of this study, blueberry extract daily therapy with dose of 1500 mg/kg BW of 7 and 14 days is shown to alleviate IL-6 expression and glomerular sclerosis scoring.
