Salsabila, Dhea (2021) Studi Antibodi Poliklonal Sebagai Kandidat Kit Deteksi African Swine Fever Berbasis Protein Rekombinan p22-ASFV Pada Hewan Coba Kelinci (Oryctolagus cuniculus). Sarjana thesis, Universitas Brawijaya.
Abstract
English Abstract
African swine fever is a disease caused by African swine fever virus (ASFV) which has a high mortality rate in pigs. ASF is a global threat due to the current lack of vaccines and the unavailability of treatment for diseases that have a morbidity and case fatality rate of up to 100% when pathogenic ASFV strains are involved. The purpose of this research is to study the character of polyclonal antibody specific results of the induction of p22-ASFV protein fragments were cloned and expressed on the cell culture media E.coli BL21 on male New Zealand White rabbits (Oryctolagus cuniculus). Samples P22-ASFV recombinant protein is derived from PUSVETMA Surabaya. Purification of the recombinant P22 protein was carried out using the SDS-PAGE electrophoresis method and continued with the electroelution method for proteins with a molecular weight of 22 kDa. Immunization of male New Zealand White rabbits (Oryctolagus cuniculus) was administered subcutaneously 800 µL then added with adjuvant CFA (Complete Freund's Adjuvant) and IFA (Incomplete Freund's Adjuvant). The Indirect ELISA method was used to measure the antibody titers and characteristics of antibodies was conducted by Western Blot method. The immunogenicity test for p22 protein using the Indirect ELISA method had the highest antibody titer values in bleeding week 7 with a value of 1.144. The Western Blot test showed that the anti-p22 antibody could recognize the p22 antigen at a molecular weight of 21,93 kDa
