Abstract

Peningkatan resistensi bakteri terhadap antibiotik merupakan salah satu masalah terbesar pada bidang kesehatan diseluruh dunia. Selain itu, pada bidang pangan, penggunaan zat antibakteri sintetis diatur dalam peraturan ketat karena berpotensi membahayakan kesehatan manusia. Maka dari itu perlu dilakukan penemuan serta pengembangan zat antibakteri baru dari bahan alami yang berspektrum luas dan aman bagi kesehatan seperti ekstrak Sargassum polycystum yang diketahui mengandung senyawa bioaktif yang bersifat antibakteri. Escherichia coli dan Staphylococcus merupakan dua jenis bakteri yang berbeda yang biasa digunakan pada penelitian untuk mengetahui seberapa luas spektrum antibakteri suatu zat. Literature review ini dilakukan dengan menggunakan metode systematic review. Tahapan pembuatannya terdiri dari penentuan topik, pencarian pustaka, pemilihan pustaka secara lebih mendalam, analisa pustaka, dan penyusunan literature review. Pustaka dicari melalui beberapa basis data seperti Google scholar dan Sciencedirect dan diutamakan dalam rentang 10 tahun terakhir atau 2011-2021. Metode ekstraksi Sargassum polycystum dapat dilakukan dengan metode konvensional dan non-konvensional. Metode konvensional seperti maserasi dan soxhlet, sedangkan metode non-konvensional seperti ultrasonik, accelerated solvent extraction (ASE), dan supercritical fluid extraction (SFE). Uji antibakteri dapat menggunakan metode difusi dan dilusi. Metode dilusi seperti dilusi padat dan cair, sedangkan metode difusi seperti cakram dan sumuran. Berdasarkan hasil beberapa penelitian, aktivitas antibakteri ekstrak Sargassum polycystum terhadap bakteri Escherichia coli diketahui ekstrak metanol Sargassum polycystum menghasilkan zona hambat tertinggi sebesar 21 mm dan dikatagorikan daya hambat sangat kuat. Konsentrasi hambat minimum (KHM) terkecil yang dihasilkan sebesar 0,25 mg/ml dengan ekstrak n-heksana, sedangkan untuk konsentrasi bakterisidal minimum (KBM) belum ada penelitian mengenai ekstrak Sargassum polycystum yang dapat membunuh 99,9% Escherichia coli. Aktivitas antibakteri ekstrak Sargassum polycystum terhadap bakteri Staphylococcus aureus dari beberapa penelitian diketahui ekstrak nheksana Sargassum polycystum menghasilkan zona hambat tertinggi sebesar 18,67 mm dan dikatagorikan daya hambat kuat. Konsentrasi hambat minimum (KHM) terendah sebesar 0,117 mg/ml yang diekstrak dengan n-hexana, sedangkan KBM terendah yang dihasilkan sebesar 0,521 mg/ml dari Sargassum polycystum yang diekstrak dengan n-hexana

English Abstract

The increase in bacterial resistance to antibiotics is one of the biggest health problems worldwide. In the food sector, the use of synthetic antibacterial substances is regulated under strict regulations because it has the potential to harm human health. Therefore, it is necessary to discover and develop new antibacterial substances from natural ingredients that have a broad spectrum and are safe for human health, such as Sargassum polycystum extract which is known to contain antibacterial bioactive compounds. Escherichia coli and Staphylococcus aureus are two different types of bacteria that are commonly used in research to determine how broad the antibacterial spectrum of a substance is. This literature review was conducted using a systematic review method. The stages of making it consist of determining the topic, searching the references, selecting the references in more depth, analyzing the literature, and compiling a literature review. References are searched through several databases such as Google scholar and Sciencedirect and are prioritized in the last 10 years or 2011- 2021. Sargassum polycystum extraction method can be done by conventional and non-conventional methods. Conventional methods such as maceration and soxhlet, while non-conventional methods such as ultrasonic, accelerated solvent extraction (ASE), and supercritical fluid extraction (SFE). Antibacterial test can be used by diffusion and dilution method. The dilution method is like broth dilution and agar dilution, while the diffusion method is like disc diffusion and well diffusion. Based on the results of several studies, the antibacterial activity of Sargassum polycystum extract against Escherichia coli bacteria is known to have Sargassum polycystum methanol extract produces the highest inhibition zone of 21 mm and is categorized as very strong. The smallest minimum inhibitory concentration (MIC) produced was 0,25 mg/ml with n-hexane extract, while for the minimum bactericidal concentration (MIC) there was no research on Sargassum polycystum extract that could kill 99,9% of Escherichia coli. Antibacterial activity of Sargassum polycystum extract against Staphylococcus aureus bacteria from several studies known that Sargassum polycystum n-hexane extract produced the highest inhibition zone of 18,67 mm and categorized as strong inhibitory. The minimum inhibitory concentration (MIC) was 0,117 mg/ml extracted with n-hexane, while the lowest concentration of minimum bactericidal concentration (MBC) was 0,521 mg/ml from Sargassum polycystum extracted with n-hexane
