Abstract

"Preeclampsia is a disorder that occurs in pregnancy, characterized by hypertension and proteinuria. Oxidative stress is thought to have an important role in the development of preeclampsia. On preeclampsia, it is suspected that there is an increase in ROS and a decrease in antioxidants. Oxidative stress can be assessed by measuring levels of Malondialdehyde (MDA), the end product of lipid peroxide. Superoxide dismutase (SOD) as a first-line antioxidant is thought to decrease in preeclampsia. The prevention and therapy strategies for preeclampsia are still very limited. Pravastatin is suspected to have antioxidant effects that can improve oxidative stress conditions that occur in preeclampsia through upregulation of endogenous antioxidant pathways. The study has aim to know the effect of pravastatin on serum MDA levels and serum SOD levels on preeclampsia rat models. Preeclampsia rat model is made by injecting N‑nitro‑L‑arginine methyl ester (L‑NAME). L-NAME is an inhibitor of nitric oxide synthase (NOS) and inhibition of NOS can increase blood pressure and vasoconstriction. The study uses a true experimental - post test only control group design. This study is consisted of 5 groups; negative control, positive control (preeclampsia rat model by induced of administration of L-NAME at a dose of 125 mg/KgBW/day since gestational age 13-19 days), treatment groups 1, 2 and 3 (Preeclampsia rat model is given pravastatin with 3 doses different, 2 mg/day, 4 mg/day and 8 mg/day) at 13-19 days of gestation. The results show that pravastatin had an effect on decreasing MDA serum levels (p=0.001) and increasing SOD serum levels (p=0.024). The increasing SOD serum level is as long with the increasing pravastatin dose. Pravastatin is suspected to be able to improve oxidative stress conditions by increasing the production of first-line antioxidants and decreasing the production of ROS. Pravastatin can reduce MDA levels and increase SOD levels in preeclampsia rat models. "
