Abstract
Penelitian ini bertujuan mengkaji senyawa fitokimia, kadar fenolik dan flavonoid total, aktivitas anti-oksidan, dan gugus fungsi pada gabah beras merah Jawa Timur yang diduga berfungsi sebagai anti-oksidan. Tiga varietas gabah beras merah, Menthik Wangi, Aek Sibundong, dan Blambangan diekstraksi menggunakan pelarut metanol. Senyawa fitokimia diuji secara kualitatif dan kadar fenolik serta flavonoid total diukur menggunakan spektrofotometer UV-Vis. Aktivitas anti-oksidan ditentukan dengan menggunakan uji FerricReducing Antioxidant Power (FRAP) dan gugus fungsional ekstrak gabah diidentifikasi dengan menggunakan FTIR. Hasil uji fitokimia menunjukkan pada gabah beras merah Menthik Wangi, Aek Sibundong, dan Blambangan terkandung senyawa fitokimia flavonoid, tritepenoid, fenolik, tannin, dan glikosida. Selain itu, pada gabah Aek Sibundong juga teridentifikasi senyawa steroid. Kadar fenolik dan flavonoid total tertinggi terdapat pada gabah beras merah Menthik Wangi, yaitu sebesar 2200,65 mg GAE/g sampel dan 1466,48 mg QE/g sampel. Aktivitas anti-oksidan tertinggi pada gabah beras merah Blambangan dengan nilai IC50 sebesar 1,09 µg/mL. Berdasarkan spektrum FTIR, teridentifikasi beberapa gugus fungsi yang ada di ketiga gabah beras merah, yaitu gugus hidroksil (OH), gugus alkana (C-H alifatik dan C-H stretching), gugus aromatik (C=O stretching), gugus alkena (C=C stretching), ikatan CH2 dan CH3, ikatan C-H aromatik dan karbonil-karbonat, gugus alkohol (CHOH stretching), dan ikatan C-O. Kesimpulan dari penelitian ini menunjukkan bahwa masing-masing gabah beras merah memiliki spesifitas kandungan fitokimia dan fungsi sebagai anti-oksidan.
English Abstract
This study investigates phytochemical compounds, total phenolic and flavonoid compound, anti-oxidant activity, and functional groups in red rice bran East Java which predicted to be an anti-oxidant. Three varieties of red rice bran, namely Menthik Wangi, Aek Sibundong, and Blambangan were extracted using methanol. Phytochemical compounds were determined qualitatively and the total phenolic and flavonoid compund were measured using a UV-Vis spectrophotometer. Anti-oxidant activity was determined using the Ferric-Reducing Antioxidant Power (FRAP) test and the functional groups of the bran extract were identified using FTIR. The results of the phytochemical test showed that red rice bran of Menthik Wangi, Aek Sibundong, and Blambangan contained flavonoid, tritepenoid, phenolic, tannin, and glycoside compounds. Moreover, red rice bran of Aek Sibundong also contains steroid compound. The highest total phenolic and flavonoid content were found in Menthik Wangi which was 2200,65 mg GAE/g sample and 1466,48 mg QE/g sample. The highest anti-oxidant activity was found in Blambangan red rice bran with an IC50 value of 1,09 µg/mL. Based on the FTIR spectrum, functional groups were identified in red rice bran, namely the hydroxyl group (OH), alkane group (CH aliphatic and CH stretching), aromatic group (C=O stretching), alkene group (C=C stretching), CH2 and CH3 bonds, aromatic and carbonyl-carbonate CH bonds, alcohol groups (CHOH stretching), and C-O bonds. The conclusion of this study showed that each red rice bran has specific phytochemical content and functions as an anti-oxidant.
