Abstract
"Hiperglikemia merupakan kondisi kadar glukosa dalam plasma darah melebihi batas normal yang merupakan tanda diabetes melitus. Kadar glukosa darah yang tidak terkontrol pada diabetes melitus dapat menyebabkan komplikasi berupa diabetes nefropati. Daun insulin (Tithonia diversifolia) memiliki kandungan flavonoid dan seskuiterpen yang mampu menurunkan kadar glukosa darah dan menangkap radikal bebas penyebab diabetes nefropati. Salah satu biomarker dari diabetes nefropati adalah kenaikan kadar serum kreatinin. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak daun insulin terhadap kadar serum kreatinin pada tikus model diabetes melitus. Penelitian ini menggunakan 36 ekor tikus Wistar jantan yang dibagi menjadi 6 kelompok, yaitu KN (tidak diberi perlakuan), kelompok yang hanya diinduksi streptozotosin (STZ) 40 mg/kgBB di antaranya KP (tanpa diberi terapi), P1, P2, dan P3 yang masing-masing diberi ekstrak daun insulin 50, 100, dan 150 mg/kgBB, serta P4 (glikuidon 10 mg/kgBB). Penelitian ini dilakukan selama 42 hari. Data yang diperoleh kemudian dianalisis menggunakan uji ANOVA atau Kruskall Wallis yang dilanjutkan dengan analisis lanjutan berupa uji Tukey HSD atau Mann Whitney. Hasil penelitian menunjukkan kelompok terapi ekstrak daun insulin 100 dan 150 mg/kgBB serta glikuidon 10 mg/kgBB memiliki perbedaan lebih rendah yang signifikan (p < 0,05) terhadap kelompok kontrol dan terapi esktrak daun insulin 50 mg/kgBB. Namun, kadar serum kreatinin baik pada kelompok kontrol maupun kelompok perlakuan masih termasuk dalam rentang normal. English Abstract
Hyperglycemia is condition of glucose levels in blood plasma exceeding normal limits which is a sign of diabetes mellitus. Uncontrolled blood glucose levels in diabetes mellitus can cause complications such as diabetic nephropathy. Insulin leaves (Tithonia diversifolia) contain flavonoids and sesquiterpenes that can reduce blood glucose levels and capture free radicals that cause diabetic nephropathy. One of the biomarkers of diabetic nephropathy is an increase in serum creatinine levels. This study aims to determine the effect of insulin leaf extract on serum creatinine levels in diabetic mellitus rats. This study used 36 male Wistar rats which were divided into 6 groups, namely KN (without treatment), then the group induced by streptozotocin (STZ) 40 mg/kgBW including KP (without therapy); P1, P2, and P3 were given insulin leaf extract 50, 100, and 150 mg/kgBW, respectively; and P4 (gliquidone 10 mg/kgBW). This research was conducted for 42 days. Data were obtained and analyzed using ANOVA or Kruskall Wallis test, followed by further analysis Tukey HSD or Mann Whitney test. The results showed that the insulin leaf extract therapy group 100 and 150 mg/kgBW and gliquidone 10 mg/kgBW had a significantly lower difference (p < 0.05) compared to the control group and the 50 mg/kgBW insulin leaf extract therapy group. However, serum creatinine levels in both the control and treatment groups were still in the normal range.
