Abstract
Spons Thorectidae sp. merupakan invertebrata multiselular sederhana yang banyak ditemukan diperairan indonesia khususnya di Kepulauan Kangean. Beberapa senyawa metabolit sekunder telah ditemukan terkandung di dalam spons dan berpotensi dalam penyembuhan penyakit kanker. Namun, perkembangan penelitian terhadap spons tersebut masih belum banyak dilakukan. Sehingga perlu dilakukan penelitian lebih lanjut untuk mengetahui potensi dari spons tersebut. Pada penelitian ini, metode ekstraksi dilakukan menggunakan metode ekstraksi sonikasi. Kemudian, uji toksisitas secara in vivo menggunakan metode Brine shrimp Lethality Test dengan menggunakan larva udang Artemia salina Leach dan uji antioksidan secara in vitro menggunakan metode uji DPPH. Hasil dari uji toksisitas dan antioksidan didapatkan bahwa ekstrak n-butanol memiliki tingkat toksisitas terbaik dengan nilai LC50 sebesar 72 ppm dan ekstrak etil asetat memiliki tingkat antioksidan terbaik dengan nilai IC50 sebesar 67,15 ppm. Hasil terbaik dari uji antioksidan akan dianalisis senyawa penyusunnya dengan LC-MS. Berdasarkan hasil LC-MS dipilih sepuluh senyawa yang memiliki potensi mengurangi ROS dan berdasarkan hasil molecular docking didapatkan bahwa Citrinin memiliki aktivitas penghambatan sel kanker terbaik terhadap reseptor 3E6I (enzim sitokrom). 6-(4-Isobutylphenyl)- 3,7 -dimethyl -7H -[1,2,4] triazolo [3,4-b] [1,3,4] thiadiazine memiliki aktivitas penghambatan sel kanker terbaik terhadap reseptor 4AOS (monooksigenase). 6,12 Dihydrobenzo [b]chromeno[4,3-e] [1,4]thiazin-6-one memiliki aktivitas penghambatan sel kanker terbaik terhadap reseptor 1OV3 (NADPH oksidase). DL-Stachydrine memiliki aktivitas penghambatan sel kanker terbaik terhadap reseptor 3PGH (siklooksigenase).
English Abstract
Spons Thorectidae sp. is a simple multi-cellular invertebrate found in Indonesia, especially in the Kangean Island. Some of secondary metabolites compounds have been found contained within the Spons and are potentially in the cure of cancer disease. However, the development of the research on the Spons is still not much done. So it is necessary to do further research to know the potential of the Spons. In this study, extraction methods were carried out using sonication extraction methods. Then, in vivo toxicity tests using the Brine shrimp Lethality Test by using the shrimp larvae of Artemia salina Leach and in vitro antioxidant tests using the DPPH test method. The results of toxicity and antioxidant tests were obtained that n-butanol extract had the best level of toxicity with a LC50 value of 72 ppm and ethyl acetate extract had the best antioxidant level with a IC50 value of 67.15 ppm. The best result of the antioxidant test will be analyzed for its constituent compounds with LC-MS. Based on the results of the LC-MS selected ten compounds that have the potential to reduce ROS and based on the results of the molecular docking acquired that Citrinin has the best inhibitory activity of cancer cells against 3E6I receptors (cytochrome enzymes). 6-(4-isobutylphenyl)-3.7-dimethyl-7H-[1, 2, 4] triazolo [3.4-B] [1, 3, 4] thiadiazine has the best cancer inhibitory activity against the 4AOS receptor (monooxygenase). 6.12 dihydrobenzo [b] chromeno [4.3-E] [1, 4] thiazin-6-one has the best inhibitory activity of cancer cells against 1OV3 receptor (NADPH oxidase). DL-Stachydrine has the best cancer inhibitory activity against the 3PGH receptor (cyclooxygenase).
