Abstract

Proteus mirabilis adalah bakteri Gram negatif yang terdapat di saluran pencernaan manusia dan hewan, namun terdapat beberapa galur Proteus mirabilis yang berpotensi menyebabkan infeksi kepada manusia. Antibiotik telah ditemukan dan dikembangkan untuk memberantas infeksi yang disebabkan oleh Proteus mirabilis tetapi dalam perjalanan waktu galur bakteri berkembang menjadi resisten terhadap antibiotik. Oleh karena itu, mengembangkan pengobatan alternatif dari tanaman obat sangat penting. Salah satu tanaman obat alami yang diduga memiliki efek antibakteri terhadap bakteri Proteus mirabilis adalah daun jarak pagar (Jatropha curcas), karena mengandung senyawa aktif seperti flavonoid, tanin, dan saponin yang disebut memiliki efek antibakteri. Tujuan dari penelitian ini adalah untuk membuktikan efek antibakteri ekstrak etanol daun jarak pagar terhadap Proteus mirabilis sehingga dapat dijadikan pengobatan infeksi yang disebabkan Proteus mirabilis. Metode yang digunakan untuk penelitian ini adalah metode dilusi agar dengan empat kali pengulangan dengan empat isolat Proteus mirabilis yang berbeda menggunakan berbagai konsentrasi ekstrak etanol daun jarak pagar yaitu 0% v/v , 1.2% v/v, 1.4% v/v, 1.6% v/v, 1.8%v/v dan 2.0% v/v terhadap Proteus mirabilis. Hasilnya menunjukkan bahwa tidak ada pertumbuhan bakteri pada 2.0% v/v. Hasil penelitian menunjukkan bahwa ada perbedaan derajat pertumbuhan bakteri antara berbagai konsentrasi ekstrak (Kruskal-Wallis, p<0,001) dan ada hubungan antara berbagai konsentrasi ekstrak dengan derajat pertumbuhan bakteri (korelasi Spearman, r = -0,985, p < 0,001). Berdasarkan hasil penelitian, dapat disimpulkan bahwa esktrak etanol daun jarak pagar memiliki efek antibakteri terhadap Proteus mirabilis secara in vitro menggunakan metode dilusi agar dengan KHM 2.0% v/v.

English Abstract

Proteus mirabilis is a Gram negative bacteria found in the digestive tract of humans and animals, but there are several strains of Proteus mirabilis that have the potential to cause infection in humans. Antibiotics have been found and developed to eradicate infections caused by Proteus mirabilis but in the course of time the strains of bacteria develop to become resistant to antibiotics. Therefore, developing alternative medicine from medicinal plants is very important. One of the natural medicinal plants which is thought to have an antibacterial effect is Jatropha leaves (Jatropha curcas), because it contains active compounds such as flavonoids, tannins, and saponins which are said to have an antibacterial effect on Proteus mirabilis. The purpose of this study is to prove the antibacterial effect ethanol extract Jatropha leaves on Proteus mirabilis so that it can be used as a treatment for infections caused by Proteus mirabilis. The method used for this research is the agar dilution method with four repetitions and with four different Proteus mirabilis isolate using various concentrations of Jatropha leaf ethanol extract, namely 0% v/v, 1.2% v/v, 1.4% v/v, 1.6% v/v, 1.8% v/v and 2.0% v/v against Proteus mirabilis. The results showed that there was no bacterial growth at 2.0% v/v. The results showed that there were differences in the degree of bacterial growth between the various extract concentrations (Kruskal-Wallis, p < 0,001) and there was a relationship between the various extract concentrations with the degree of bacterial growth (Spearman correlation, r = -0.985, p < 0,001). Based on the results of the study, it can be concluded that the ethanol extract of Jatropha leaves has an antibacterial effect on Proteus mirabilis in vitro using the agar dilution method with 2.0% v/v MIC.
