Abstract

Semanggi air merupakan tanaman obat yang memiliki komponen fitokimia flavonoid, alkaloid, steroid, fenol, tanin, minyak atsiri dan saponin. Penelitian ini dilakukan untuk mengetahui efek pemberian ekstrak ethanol semanggi air terhadap histologi hepar dan ginjal. Penelitian ini menggunakan 24 tikus betina (Rattus norvegicus) berumur 2-3 bulan dengan berat 130-150 g. Penelitian ini menggunakan rancangan acak lengkap yang terdiri dari empat group. K0 (kontrol) adalah kelompok tanpa pemberian ekstrak ethanol semanggi air. P1,P2, dan P3 adalah kelompok yang diberi ekstrak ethanol semanggi air dengan masing-masing dosis 0,216 mg/gBB, 0,432 mg/gBB dan 0,648 mg/gBB. Ekstrak ethanol semanggi air diberikan secara oral selama 15 hari, dimulai pada fase estrus. Pada hari ke 16, tikus dibedah dan diisolasi organ hepar dan ginjal. Hepar dan ginjal difiksasid engan etanol selama 24 jam kemudian dibuat histologi dengan metode parafin dengan pewarnaan hematoxylin eosin. Struktur jaringan histologi hepar dan ginjal dianalisis dengan menghitung jumlah nekrosis (piknosis dan kariolisis) hepatosit dan sel tubular ginjal. Data dianalisis dengan One-way ANOVA. Hasil menunjukkan bahwa pemberian ekstrak etanol semanggi air tidak meningkatkan nekrosis hepatosit tetapi meningkatkan nekrosis sel tubular ginjal.

English Abstract

Water clover is a medicinal plant that has phytochemical components such as flavonoids, alkaloids, steroids, phenols, tannins, essential oils, and saponins. This research was conducted to observe the effect of administering water clover ethanol extract on liver and kidney histology. This study used 24 female rats, which has 2-3 months old with 130 – 150 g of weight. The design used in this study is completely randomized design which consist of four gropus. K0 (control) is a group that is not given ethanol extract of water clover. P1, P2 and P3 were the groups that given water clover ethanol extract with doses of 0.216 mg / gBB, 0.432 mg / gBB and 0.648 mg / gBB respectively. The water clover ethanol extract were given orally for 15 days, starting in the estrous phase. On the 16th day, the rat were sacrified, liver and kidney were collected. The liver and kidney was fixed with ethanol for 24 hours, then made the histology preparation by paraffin method with hematoxylin eosin staining. The histological structure of the liver and kidney tissue was analyzed by counting the number of necrosis (pycnosis and karyolysis) hepatocytes and renal tubular cells. The data were analyzed by One-Way ANOVA. The results showed that administration of ethanol extract of water clover did not increase hepatocyte necrosis, but increased necrosis of renal tubular cells
