Abstract

Ulkus lambung merupakan kondisi patologis lambung akibat rusaknya lapisan mukosa hingga submukosa dari lambung yang dapat menimbulkan gejala nyeri,perdarahan. Ulkus lambung umumnya disebabkan oleh pemakaian Non Steroid Inflammation Drug (NSAID) dan infeksi Helicobacter pylori. NSAID bekerja melalui hambatan jalur COX-1 dan COX-2 yang menyebabkan hilangnya lapisan pelindung lambung dan inflamasi mengakibatkan timbulnya ulkus lambung. Daun kemangi diketahui memiliki efek antiinflamasi dan antioksidan oleh bahan aktif flavonoid melalui hambatan mediator inflamasi dan pencegahan produksi Reactive Oxygen Species (ROS) yang mampu mengurangi keparahan ulkus lambung. Penelitian ini dilakukan untuk mengetahui pengaruh ekstrak ethanol daun kemangi Ocimum africanum terhadap skor keparahan ulkus lambung tikus induksi indometasin secara makroskopis. Penelitian ini menggunakan 25 tikus Rattus novergicus model ulkus lambung dengan induksi indometasin yang dibagi menjadi 5 kelompok terdiri dari kontrol negatif, kontrol positif, perlakuan satu, perlakuan dua, dan perlakuan tiga. Kelompok kontrol positif dan perlakuan diinduksi indometasin (30 mg/KgBB per oral. Kemudian, diberikan ekstrak ethanol ke kelompok perlakuan secara berurutan 150 mg/kgBB, 300 mg/kgBB, dan 600 mg/kgBB selama 1 hari setiap 8 jam. Kemudian hewan coba dikorbankan dan dinilai secara makroskopis menggunakan skor keparahan ulkus lambung gusdinar. Hasil uji beda Mann Whitney menunjukkan terdapat perbedaan signifikan dengan p = 0,005 (p = < 0,05) dan uji korelasi spearman menunjukan korelasi negatif kuat dengan p = 0,008 (p = <0,05). Oleh karena itu, dapat disimpulkan terdapat pengaruh ekstrak ethanol daun kemangi (Ocimum africanum) terhadap skor keparahan ulkus lambung.

English Abstract

Gastric ulcer is a pathological condition of the stomach due to damage to the mucosal lining to the submucosa of the stomach which can cause symptoms of pain, bleeding. Gastric ulcers are generally caused by the use of Non-Steroid Inflammation Drug (NSAID) and Helicobacter pylori infection. NSAIDs work through the inhibition of COX-1 and COX-2 pathways that cause the loss of the protective lining of the stomach and cause gastric ulcers. Basil leaves are known to have anti-inflammatory and antioxidant effects by the active ingredient of flavonoid through inhibition of inflammatory mediators and decrease of Reactive Oxygen Specimen (ROS) that can reduce the severity of gastric ulcers. This research was conducted to determine the effect of basil leaves (Ocimum africanum) ethanolic extract on the severity score of indometacin induced gastric ulcer mice. This study used 25 Rattus novergicus mice which were modeled gastric ulcer through indomethacin induction which were divided into 5 groups consisting of negative controls, positive controls, one treatment, two treatments, and three treatments. The positive control group and treatment groups were given indomethacin 30 mg/KgBB orally. Then, ethanol extract was given to the treatment groups respectively 150 mg/kgBW, 300 mg/kgBW, and 600 mg/kgBW for 1 day every 8 hours. Then the rats were sacrificed and assessed macroscopically using the gusdinar gastric ulcer severity score The Mann Whitney test showed a significant influence p = 0.005 (p = <0.05) and Spearman correlation test showed significant negative correlation p = 0.008 (p = <0.05). It can be concluded that there is an effect of ethanolic extract of basil leaves (Ocimum africanum) toward the severity score of gastric ulcer.
