Abstract

Sapi perah pejantan unggul digunakan sebagai sumber koleksi sperma yang dapat digunakan untuk Inseminasi Buatan (IB) ataupun Transfer Embrio (TE). Penentuan keberhasilan dalam IB dan TE dipengaruhi oleh kualitas sperma yang baik. Tujuan dari penelitian ini adalah mengetahui fertilitas pejantan yang akan digunakan sebagai pembuatan semen beku dengan mengidentifikasi ada tidaknya korelasi marka fertilitas spesifik pada sapi perah FH jantan (osteopontin) menggunakan sampel serum terhadap level testosteron. Penelitian menggunakan 12 sampel Sapi Pejantan FH yaitu 6 sampel diperoleh dari Balai Besar Inseminasi Buatan (BBIB) Singosari Malang yang merupakan sampel kualitas unggul dan 6 sampel diperoleh dari Peternakan Rakyat yang merupakan sampel kualitas biasa. Analisa profil pita protein osteopontin dilakukan dengan metode Sodium Dodecyl Sulphate Poly Acrylamide Gel Electrophoresis (SDS-PAGE) dan analisa kadar testosteron dilakukan dengan metode Enzyme-Linked Immunosorbent Assay (ELISA). Gel SDS PAGE divisualisasikan dan diperoleh hasil protein osteopontin dengan berat molekul 62,0-70,7 kDa. Sementara hasil Uji ELISA Hormon Testosteron sampel Sapi Kualitas Unggul sebesar 363,4762 μg/dL dan sampel Sapi Kualitas biasa sebesar 144 μg/dL. Identifikasi korelasi dilakukan menggunakan Uji Spearman pada software R Studio sehingga didapatkan hasil adanya korelasi antara intensitas osteopontin serum terhadap kadar hormon testosteron pada Sapi Pejantan FH dan intensitas osteopontin serum mempengaruhi tingkat fertilitas pada Sapi Pejantan FH.

English Abstract

Superior dairy cows are used as a source of sperm collection that can be used for Artificial Insemination (AI) or Embryo Transfer (ET). Determination of success in AI and ET is approved by good sperm quality. Fertility in dairy cows is controlled by hormone testosterone and also can be seen through osteopontin levels in semen. The purpose of this study is to determine the fertility of males that will be used to make frozen semen, to learn about serum osteopontin concentrations against testosterone levels in superior FH dairy cows and to find out the profile of serum osteopontin protein bands in superior FH dairy cows. This study used 12 FH bulls samples, 6 of it were obtained from the Singosari Center for Artificial Insemination (BBIB) in Malang, which quality is considered superior and 6 samples obtained from People's Farms which quality is considered ordinary. Analysis of the osteopontin protein band profile was performed using the Sodium Dodecyl Sulphate Poly Acrylamide Gel Electrophoresis (SDS-PAGE) method and analysis of testosterone levels was carried out using the Enzyme-Linked Immunosorbent Assay (ELISA) method. SDS PAGE Gel was visualized and the result showed proteins with molecular weights of 62.0-70.7 kDa. While the results of the ELISA Test for Hormone Testosterone in Superior Quality Cow samples were 363.4762 μg / dL and the ordinary Quality Cattle samples were 144 μg/dL. The correlation identification was carried out using the Spearman Test in R Studio software so that the correlation between serum osteopontin intensity and testosterone levels in FH Male Cow and serum osteopontin intensity affect fertility level in FH Male Cows.
