Abstract

Reseptor FSH yang diekspresikan pada membran plasma sel Sertoli hewan jantan dan sel Granulosa hewan betina sangat penting untuk menerima FSH yang diperlukan dalam regulasi sistem reproduksi. Adanya polimorfisme pada gen RFSH berpotensi untuk mempengaruhi sistem reproduksi dan performa hewan secara umum. Penelitian ini bertujuan untuk mengidentifikasi polimorfisme gen RFSH terhadap performa pejantan yang meliputi lingkar skrotum, BCS, dan konsentrasi spermatozoa pejantan sapi perah FH. Sampel berupa darah diambil dari 13 sapi FH di BBIB Singosari dan dianalisa di Laboratorium Animal Disease Diagnosis Fakultas Kedokteran Hewan Universitas Brawijaya dan Laboratorium Jurusan Biologi Fakultas Sains dan Teknologi UIN Maulana Malik Ibrahim Malang. Teknik yang digunakan untuk mengidentifikasi polimorfisme pada RFSH adalah PCR-RFLP. Metode yang digunakan untuk melihat korelasi antara polimorfisme dan performa pejantan adalah Spearman’s Rank Correlation Coefficient. Hasil yang didapat adalah adanya korelasi sedang dan searah antara tingkat kemunculan polimorfisme G>T dengan BCS (rho = 0.5387884) dan lingkar skrotum (rho = 0.4269932) serta korelasi lemah dan berlawanan arah dengan konsentrasi spermatozoa (rho = -0.3501423) namun secara statistik korelasi tersebut tidak signifikan (p > 0,05).

English Abstract

FSH receptor in the plasmic membrane of male's Sertoli cell and female's Granulosa cell is important for receiving FSH which is needed in regulating the reproductive system. Polymorphism in RFSH gene can potentially influence reproduction and cattle performance in general. The goal of this research is to identify RFSH gene polymorphism influence on male's performance of PFH dairy cows, including testes circumferences, BCS, and sperm concentration. Blood samples are collected from 13 FH dairy cows in BBIB Singosari and analyzed in Animal Disease Diagnosis Laboratory of Veterinary Faculty, University of Brawijaya and Laboratory of Biology Course, Science and Technology Faculty, UIN Maulana Malik Ibrahim Malang. Polymorphism of RFSH was identified by using PCR-RFLP. Correlation between polymorphisms and performance was determined using Spearman’s Rank Correlation Coefficient. The result showed that there was an intermediate correlation between G>T polymorphism and BCS (rho = 0.5387884) and testes circumferences (rho = 0.4269932) in the same direction. There was a weak correlation with the opposing direction between G>T polymorphism and sperm concentration (rho = -0.3501423), but all of those correlations were statistically insignificant (p > 0,05).
