Abstract
Pseudomonas aeruginosa merupakan salah satu patogen penyebab infeksi nosokomial dengan kasus resistensi antibiotika yang tinggi. Resistensi bakteri tersebut diakibatkan proses pembentukan biofilm. Daun kelor (Moringa oleifera) sebagai bahan pengganti obat yang mengandung zat aktif tannin, flavonoid, dan alkaloid sebagai antibiofilm melalui mekanisme pencegahan quorum sensing pada bakteri Pseudomonas aeruginosa . Tujuan?penelitian?ini adalah?untuk?mengetahui potensi ekstrak?etanol daun kelor sebagai penghambat pembentukan biofilm Pseudomonas aeruginosa secara in vitro. Penelitian ini?menggunakan metode?tabung. Terdapat 7 kelompok dengan?6 kelompok perlakuan dengan konsentrasi 1,675%, 3,125%, 6,25%, 12,5%, 25%, 50%, kontrol negatif berisi ekstrak konsentrasi 0%. Dengan pengulangan dan perapatan sebanyak 4 kali dengan konsentrasi 10%, 12,5%, 15%, 17,5%, 20%, 22,5%, 25%. Hasil penelitian dinilai dengan cara pengamatan secara langsung dan uji statistic nonparametrik meliputi Kolmogorov-Smirnov dan Shapiro-Wik, Kruskall Wallis, uji.Mann-Whitney, dan uji?korelasi Spearman. Hasil?penelitian?menunjukkan bahwa, secara visual terlihat penipisan cincin biofilm pada tabung. Hasilianalisis uji KruskaliWallis.didapatkan p=0,000.(p<0,05). Hasil uji Spearman yaitu 0,000. Kesimpulan3penelitian3ini3adalah3ekstrak etanol daun kelor dapat3menghambat?pembentukan?biofilm?pada bakteri Pseudomonas aeruginosa secara in vitro dengan signfikan.
English Abstract
Pseudomonas aeruginosa is one of a pathogen that causes common cases of nosocomial infection with antibiotic resistance. This resistance bacteria happens due to process of biofilm forming. Kelor leaf (Moringa oleifera) as the substitute of medicine in which consists of active substances such as tannin, flavonoid and alkaloid as antibiofilm through mechanism prevention quorum sensing in bacteria Pseudomonas aeruginosa. The objective of this study is to learn the potential benefit of extract of etanol in kelor leaf as a resistor in forming biofilm Pseudomonas aeruginosa in vitro. This study is using tube test method. There are 7 groups with 6 groups concentration treatment 1,675%, 3,125%, 6,25%, 12,5%, 25%, 50%, negative control consist of concentration extract 0%. With repetition and sealing as many as 4 times with concentration 10%, 12,5%, 15%, 17,5%, 20%, 22,5%, 25%. The result was rated with direct observation and statistical assessment nonparametrik comprise Kolmogorov-Smirnov and Shapiro-Wik, Kruskal Wallis, assessment Mann-Whitney, and assessment Spearman correlation. The study outcome shows visually, it is seen there are depletion on biofilm ring in tube. The outcome of Kruskal Wallis assessment is p=0,000 (p<0,05). The outcome of Spearman is 0,000. The finding of this study is etanol extract on kelor leaf can be resistor in forming biofilm on Pseudomonas aeruginosa bacteria in vitro significantly.
