Abstract
Streptomyces hygroscopicus (S.hygroscopicus) adalah bakteri tanah kelompok Gram positif memiliki kemampuan memproduksi metabolit sekunder hasil fermentasi yang dapat bersifat terapetik. Metabolit hasil fermentasi S.hygrocospicus sulit untuk dikembangkan sebagai sediaan obat karena masih berupa ekstrak kasar yang belum diketahui isolat senyawa aktifnya, sehingga akan sulit dalam penentuan dosis obat agar dapat memberikan efek terapi Oleh karena itu, perlu dilakukan penelitian eksplorasi untuk mempelajari metabolit sekunder dari hasil fermentasi S.hygroscopicus, berupa penyempitan senyawa metabolik yang ditarik dengan metode ekstraksi dan fraksinasi dengan pelarut etil asetat-n heksana yang kemudian akan diindentifikasi dengan KLT. Penelitian ini menggunakan metode ekstraksi dengan corong pisah dan kemudian dilakukan fraksinasi menggunakan kolom kromatografi BUCHI (Sepacore®) Flash Chromatography dan Reveleris® PREP Purification System sehingga didapatkan hasil fraksinasi sejumlah 47 dan 60 fraksi. Selanjutnya sampel-sampel tersebut dilakukan profiling dengan menggunakan KLT dan diberikan penampak noda KOH 10% dan p-Anisaldehyde – sulfuric acid sehingga didapatkan berbagai golongan senyawa yaitu Monoterpenes, Triterpenes, Steroid, Saponin, Coumarin, Scopoletin, dan Alkaloid. Gradien perbandingan pelarut etil asetat dan n-heksana yang secara optimal menghasilkan noda tersebut adalah 65% n-heksana : 35% etil asetat, dan 50% n-heksana : 50% etil asetat.
English Abstract
Streptomyces hygroscopicus (S.hygroscopicus) is a Gram-positive soil bacterium that can produce secondary metabolites from fermentation that have a therapeutic effect. However, the secondary metabolism of fermented S.hygrocospicus is difficult to develop as a drug product because it is still in the form of crude extracts that have not been known which compound as the active compound, which makes it be difficult in determining drug dosages to provide a therapeutic effect. Therefore, exploratory research is needed to study further about secondary metabolites from the S.hygroscopicus fermentation, in the form of narrowing the metabolic compounds drawn by extraction and fractionation methods with ethyl acetat – n-hexane solvents which will then be identified by Thin Layer Chromatography. This study used a separate funnel as the extraction method and then fractionation was done using the BUCHI (Sepacore®) Flash Chromatography and Reveleris® PREP Purification System column so that the results of the fractionation of 47 from BUCHI (Sepacore®) Flash Chromatography and 60 from Reveleris® PREP Purification System were obtained. Then the samples were profiled using Thin Layer Chromatography using reagent 10% KOH and p-Anisaldehyde - sulfuric acid was obtained to obtain various groups of compounds, namely Monoterpenes, Triterpenes, Steroids, Saponins, Coumarin, Scopoletin, and Alkaloids. The most optimal gradient were 65% n-hexane:35% ethyl acetate and 50% n-hexane:50% ethyl acetate.
