Abstract

Tujuan dari penelitian ini untuk mengevaluasi efek samping pemberian terapi self-antigen dsDNA terhadap fungsi ginjal mencit model lupus yang diinduksi pristan. 24 Balb/c betina dibagi menjadi tiga kelompok; kelompok negatif, kelompok kontrol positif, dan kelompok perlakuan. Kelompok positif dan perlakuan diinjeksikan pristan 0,5 ml intraperitoneal. Setelah 12 minggu pasca injeksi, manifestasi klinis dan serologis mencit dievaluasi. Self-antigen dsDNA ditambahkan Polyethyleneimine (PEI) diberikan pada kontrol perlakuan dengan tiga dosis berbeda setiap minggu (0.05 μg/ml, 0.5 μg/ml, 5 μg/ml). Sampel dianalisa untuk menentukan fungsi ginjal melalui kadar ureum dan kreatinin menggunakan spektofotometri. Hasil penelitian menunjukan mencit lupus induksi pristan dengan terapi self-antigen dsDNA ditambahkan PEI menggunakan dosis bertahap dapat menurunkan kadar urea secara signifikan (p=0,009) dan menurunkan kadar kreatinin (p=0,801) dibandingkan kelompok kontrol positif. Self-antigen dsDNA dengan PEI dapat menurunkan kadar ureum dan kreatinin sehingga menjadi immunotherapy yang potensial pada mencit lupus yang diinduksi pristan

English Abstract

The aim of this study was to evaluate the side effect of self-antigen dsDNA therapy on renal function in pristine-induced Lupus (PIL) mice. 24 Female Balb/c mice were devided into three different groups; negative group, positive control group, and treatment group. The positive and treatment groups were given a single intraperitoneal injection of 0.5 ml pristane. Starting at 12 weeks after injection, the mice were evaluated for Lupus clinical and serological manifestations. Self-antigen dsDNA complexed with polyethyleneimine (PEI) was administered to the treatment group once every week with three different doses (0.05 μg/ml, 0.5 μg/ml, 5 μg/ml). Samples were analyzed to determine renal function by evaluating urea and creatinine levels using spectophotometry. Analysis for side effect showed that PIL treated with escalating dose self antigen dsDNA significantly decrease urea (p=0.009) and not significantly decrease creatinine (p=0,801) compared to positive control. Self-antigen dsDNA coupled with PEI was able to decrease urea and creatinin levels, thus potentialy be a safe immunotherapy againts pristine-induced SLE Balb/c mice.
