Abstract

Jeruk JC (Citrus limonia. Osbeck) merupakan varietas jeruk batang bawah yang tahan terhadap stress kekeringan namun mudah terinfeksi penyakit kanker jeruk yang disebabkan oleh Xanthomonas axonopodis pv.citri (XAC). Penelitian ini bertujuan untuk mengetahui potensi bakteri endofit jeruk JC dalam menghambat pertumbuhan XAC secara in vitro dan in vivo serta mengidentifikasi bakteri endofit unggul berdasarkan sekuen 16S rDNA. Bakteri endofit diisolasi dengan serial dilusi dan metode spread plate pada media NA. Bakteri endofit diseleksi kemampuanya dalam menghambat pertumbuhan XAC secara in vitro dengan metode agar well difussion. Metabolit sekunder dalam bentuk CFS (Cell free supernatant) diuji potensinya dalam menghambat pertumbuhan XAC secara in vitro dengan metode agar well difussion. Uji in vivo bakteri endofit terpilih dilakukan dengan mengaplikasikan suspensi sel dan CFS pada daun uji sebelum inokulasi XAC. Isolat dengan kemampuan paling baik diidentifikasi berdasarkan sekuen 16S rDNA. Sebanyak 21 isolat bakteri endofit berhasil diisolasi dan isolat B1 dan C8 memiliki potensi tinggi dalam menghambat XAC dengan diameter zona hambat secara berurutan sebesar 8,42 ± 0,12 mm dan 7,52 ± 0,1 mm. Aplikasi suspensi sel dan CFS isolat terpilih secara efektif dapat memberikan proteksi terhadap penyakit kanker jeruk dengan menurunkan insidensi penyakit hingga 28 % dengan luas gejala hingga 4,1 ± 1,25 % dan jumlah titik nekrosis mencapai 3-4 titik. Isolat B1 terindentifikasi sebagai Staphylococcus pasteuri dan isolat C8 teridenfikasi sebagai S. warneri

English Abstract

Rangpur lime (Citrus limonia. Osbeck) is a citrus cultivar that is commonly used as rootstock and known for its high resistance to drought stress but it is very susceptible to citrus canker infection caused by Xanthomonas axonopodis pv. citri (XAC). This research aims to study the potency of endophytic bacteria isolated from rangpur lime to inhibit the XAC based on in vitro and in vivo treatment and to identify selected endophytic bacteria isolate based on 16S rDNA sequence. Endophytic bacteria isolated from the citrus leaf through serial dilution technique and spread plate method on NA medium. Endophytic bacteria is screened based on its ability to inhibit the growth of XAC in vitro culture through agar well diffusion technique. Secondary metabolite in the form of cell-free supernatant (CFS) of each selected isolate is subjected to inhibition assay against XAC based on agar well diffusion method. In vivo inhibition test of selected bacteria against XAC was carried out by applying its cell suspension and CFS to healthy rangpur lime leaves before the inoculation of XAC pathogen. Selected isolates were identified based on the 16S rDNA sequence. As much as 21 bacterial isolates were isolated and 2 isolates (B1 and C8) demonstrating strong inhibition against XAC with 8.42 ± 0.12 mm dan 7.52 ± 0.1 mm inhibition zones diameter respectively. The application of cell suspension or CFS of selected bacteria decreased the disease incidence to 28 %, the leaf area with lesions to 4.1 ± 1.25 % and the necrotic spots to 3-4 spots. Isolate B1 and C8 were identified as Staphylococcus pasteuri and S. warneri respectively
