Abstract

Latar Belakang: Ekstrak etanol daun kemangi (Ocimum sanctum) memiliki efek anti jamur terhadap Candida albicans melalui penghambatan filamentasi sel ragi. Tujuan: Untuk membuktikan efek ekstrak etanol daun kemangi (Ocimum sanctum) terhadap pembentukan biofilm Candida albicans secara in vitro dan mengetahui konsentrasi hambat biofilm minimal yang diperlukan. Metode: Desain penelitian menggunakan true experimental-post test only group dengan metode tabung. 7 Konsentrasi berbeda dari ekstrak (0%, 10%, 15%, 20%, 25%, 30%, and 35%) digunakan dalam penelitian ini. Hasil bentukan cincin biofilm yang didapat, diukur secara kuantitatif menggunakan Mean Gray Value pada Adobe Photoshop CS6. Hasil: Kenaikan konsentrasi ekstrak berbanding lurus dengan penipisan cincin biofilm pada tabung. Nilai Konsentrasi Hambat Minimal Biofilm (KHBM) ekstrak etanol daun kemangi adalah konsentrasi 20%. Uji korelasi Pearson menunjukkan korelasi sangat kuat dan signifikan (r = 0,900, p=0,000), dan uji komparasi Oneway ANOVA menunjukkan adanya perbedaan nilai yang signifikan antara rerata tiap kelompok (p=0,000). Kesimpulan: ekstrak etanol Ocimum sanctum dapat menghambat pembentukan biofilm Candida albicans secara in vitro.

English Abstract

Background: Ethanol extract of basil leaves (Ocimum sanctum) has an antifungal effect on Candida albicans by inhibiting the filamentation of yeast cell. Objectives: To prove the effect of ethanol extract of basil leaves (Ocimum sanctum) on the formation of Candida albicans biofilms in vitro and determine the minimum inhibitory concentration of biofilms needed. Methods: The Design of this study is a true experimental post-test only group using tube method. 7 different extract concentrations (0%, 10%, 15%, 20%, 25%, 30%, and 35%) was used in this study. The results of biofilm ring formation obtained and measured quantitatively using Mean Gray Value in Adobe Photoshop CS6. Results: Increase in extract concentration is directly proportional to the thinning of the biofilm ring on the tube. The vaue of Minimum Biofilm Inhibitory Concentration (MBIC) is reached at 20% extract concentration. Pearson correlation test showed a very strong and significant correlation (r = 0.900, p = 0,000), and the Oneway ANOVA comparison test showed a significant difference between the mean of each group (p = 0,000). Conclusion: Ethanol extract Ocimum sanctum can inhibit the formation of Candida albicans biofilms in vitro.
