Abstract

Tingkat interleukin (IL)-6, jumlah sel T CD4+, dan jumlah sel T CD8+ pada kanker payudara lebih tinggi daripada kondisi normal. Cheral mengandung ekstrak meniran dan kunyit. Meniran mengandung asam ellagic yang dapat menurunkan tingkat IL-6. Kunyit mengandung curcumin yang dapat menurunkan IL-6, mencegah terjadinya deplesi sel T yang diinduksi oleh tumor, dan meningkatkan kemampuan sel T efektor dalam membunuh sel kanker. Cheral diharapkan dapat memberi efek anti-kanker yang optimal. Penelitian ini bertujuan untuk mengetahui pengaruh cheral terhadap jumlah relatif sel T CD4+CD62L-, CD8+CD62L-, dan makrofag CD11b+IL-6+ limpa pada mencit (Mus musculus) model kanker payudara. Mencit dibagi ke dalam tuju kelompok : normal; kanker payudara (diinjeksi dengan 7,12-dimethylbenzanthracene (DMBA)); kanker payudara dan diinjeksi dengan cisplatin; serta kanker payudara dan diberi cheral dosis 1, 2, 3, dan 4 secara oral. Mencit diaklimasi selama seminggu. Mencit diinduksi kanker payudara menggunakan DMBA sebanyak enam kali (satu kali setiap minggunya) dengan dosis 0,015 mg/g BB. Mencit diberi cheral secara oral dengan 4 dosis berbeda (dosis 1 sebesar 0,492 mg/g BB, dosis 2 sebesar 1,184 mg/g BB, dosis 3 sebesar 2,46 mg/g BB, dan dosis 4 sebesar 6,15 mg/g BB). Mencit diinjeksi secara intraperitoneal dengan cisplatin selama dua minggu (dua hari sekali) dengan dosis 0,003 mg/g BB. Organ limpa diisolasi dan diproses untuk flow cytometry. Hasil flow cytometry dianalisis menggunakan program BD CELLQUEST ProTM. Analisis statistik dilakukan menggunakan program IBM SPSS 22 uji one way ANOVA (p˂0,05), dilanjutkan dengan uji Duncan’s Multiple Range. Kesimpulan penelitian ini yaitu jumlah relatif sel T CD4+CD62L-, CD8+CD62L-, dan makrofag CD11b+IL-6+ pada mencit kanker payudara mendekati jumlah relatif sel-sel tersebut pada mencit normal secara berturut-turut saat mencit kanker payudara diberi cheral dosis 3; 1 dan 3; serta 2.

English Abstract

Breast cancer is associated with higher level of interleukin (IL)-6, CD4+, and CD8+ T cells than that of normal condition. Cheral contains chamber bitter (Phyllanthus niruri) and turmeric (Curcuma longa) extract. Chamber bitter contains ellagic acid that can reduces IL-6 level. Turmeric contains curcumin that can decrease IL-6, prevent T cells depletion that induced by tumor, and increase effector T cells ability to kill cancer cells. Cheral is expected to provide an optimal anti-cancer effect. This research aims to study the effect of cheral on spleen’s relative quantity of CD4+CD62L-, CD8+CD62L- T cells, and CD11b+IL-6+ macrophage cells in mice’ (Mus musculus) breast cancer. Mice were classified into 7 groups : normal; breast cancer (injected with 7,12-dimethylbenzanthracene (DMBA)); breast cancer and injected with cisplatin; breast cancer and administered orally with cheral dosage 1, 2, 3, and 4. Mice were acclimated for a week and then injected with (DMBA) six times (once a week) with dosage 0,015 mg/g BW. Cheral was administered orally to mice with four different dosages (dosage 1 is 0,492 mg/g BW, dosage 2 is 1,184 mg/g BW, dosage 3 is 2,46 mg/g BW, and dosage 4 is 6,15 mg/g BW). Mice were injected intraperitoneally with 0,003 mg/g BW cisplatin for two weeks (once every two days). Spleen was isolated and processed for flow cytometry. Relative quantity of CD4+CD62L-, CD8+CD62L- T cells, and CD11b+IL-6+ macrophage cells were analyzed with BD CELLQUEST ProTM program. Quantitative data were analyzed by one way ANOVA (p˂0,05) and followed by Duncan’s Multiple Range Test (using IBM SPSS 22 program). The conclusion of this research is relative number of CD4+CD62L-, CD8+CD62L- T cells, and CD11b+IL-6+ macrophage cells of breast cancer mice are similar to that of normal group while breast cancer mice were administered orally with cheral dosage 3; 1 and 3; and 2 respectively.
