Abstract

Candida albicans merupakan jamur yang dapat menginfeksi saluran genital dan rentan terjadi saat kehamilan. Daun katuk dan tapak liman mengandung flavonoid dan saponin, dan sebagai imunomodulator alami yang dapat meningkatkan respon imunitas tubuh. Sitokin proinflamasi dihasilkan oleh sel limfosit T yang telah teraktivasi dan berperan dalam respon inflamasi. Tujuan penelitian yaitu mengetahui efektivitas kombinasi ekstrak daun katuk (S. androgynus) dan tapak liman (E. scaber) terhadap produksi sitokin IFN-γ, TNF-α dan IL-4 mencit bunting terinfeksi C. albicans. Penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan tujuh perlakuan dan tiga pengulangan dalam tiga kelompok pembedahan. Mencit betina diinjeksi dengan hormon, kemudian mencit jantan dan betina dikumpulkan dalam satu kandang. Pagi hari dilakukan pengamatan vaginal plug dan pengecekan vaginal smear. Pemberian ekstrak daun dilakukan pada hari pertama kebuntingan hingga pembedahan dan infeksi C. albicans pada hari ke-5 kebuntingan secara intravagina sebanyak 40 μl. Pembedahan mencit dilakukan pada hari ke-10, ke-14 dan ke-18 kebuntingan untuk diambil organ limpa dan dianalisis flow cytometry. Analisis data menggunakan ANOVA tingkat kepercayaan 95% dan dilanjutkan uji Duncan. Hasil dari penelitian yaitu infeksi C. albicans pada mencit bunting menyebabkan peningkatan inflamasi berkaitan dengan peningkatan jumlah produksi sitokin proinflamasi (IFN-γ dan TNF-α) serta penurunan sitokin IL4 yang diproduksi oleh sel limfosit T. Pemberian kombinasi ekstrak daun katuk dan tapak liman pada dosis P3 (50% katuk dan 50% tapak liman) mampu menurunkan jumlah sitokin proinflamasi dan meningkatkan sitokin IL-4.

English Abstract

Candida albicans is a fungus that can infect the genital tract and is prone to pregnancy. Katuk and tapak liman leaves contain flavonoids and saponins, and as natural immunomodulators that can enhance the body's immune response. Proinflammatory cytokines are produced by T lymphocytes that have been activated and play a role in the inflammatory response. The aim of the study was to determine the effectiveness of the combination of katuk (S. androgynus) and tapak liman (E. scaber) leaf extract on the total of cytokines IFN-γ, TNF-α and IL-4 of pregnant mice infected C. albicans. The study used a completely randomized design (CRD) with seven treatments and three repetitions in three surgical groups. Female mice are injected with hormones, then male and female mice are collected in one cage. In the morning a vaginal plug and vaginal smear were observed. The administration of leaf extract was carried out on the first day of pregnancy until surgery and infection with C. albicans on the 5th day of pregnancy by intravagina as much as 40μl. Mice surgery was carried out on the 10th, 14th and 18th days of pregnancy for the spleen taken and flowcytometry was analyzed. Data analysis using ANOVA 95% confidence level and continued with Duncan test. The results of the study were C. albicans infection in pregnant mice causing an increase in inflammation associated with an increase in the total of proinflammatory cytokines such as IFN-γ and TNF-α and decrease in IL4 cytokines produced by T lymphocyte cells. In the P3 dose (50% katuk and 50% tapak liman) is able to reduce the number of proinflammatory cytokines and increase IL-4 cytokine.
