Abstract

Proses penyembuhan soket pasca pencabutan gigi perlu ditingkatkan untuk mencegah resorpsi berlebih dan mengembalikan fungsi tulang alveolar dalam menopang gigi geligi serta melindungi struktur penting dalam tulang. Daun tempuyung (Sonchus arvensis L.) memiliki kandungan triterpenoid. Senyawa tersebut dapat menghambat osteoklastogenesis dengan cara menghambat ekspresi gen spesifik osteoklas, yaitu Tartrate-resistance Acid Phosphatase (TRAP), sehingga pembentukan osteoklas menurun dan proses penyembuhan soket pasca pencabutan gigi akan meningkat. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak daun tempuyung (Sonchus arvensis L.) terhadap jumlah osteoklas pada penyembuhan soket pasca pencabutan gigi pada tikus putih (Rattus norvegicus). Penelitian ini dilakukan dengan metode eksperimental laboratoris secara in vivo dan membagi hewan coba ke dalam 4 kelompok, yaitu kelompok K7 dan K 14 (tidak diberi ekstrak daun tempuyung) dan P7 serta P14 (diberi ekstrak daun tempuyung), kemudian dilakukan dekaputasi pada hari ke 7 dan 14, dan dibuat sediaan histologis dengan pewarnaan HE lalu dihitung jumlah osteoklasnya. Analisis data menggunakan uji T tidak berpasangan menunjukkan perbedaan yang signifikan pada setiap 2 kelompok yang dibandingkan, kecuali pada kelompok K7 dan P7. Uji korelasi Pearson menunjukkan adanya hubungan ke arah negatif yang berarti semakin lama pemberian ekstrak daun tempuyung, jumlah osteoklas akan semakin menurun. Berdasarkan penelitian ini, dapat disimpulkan bahwa ekstrak daun tempuyung (Sonchus Arvensis L.) berpengaruh terhadap jumlah osteoklas pada penyembuhan soket pasca pencabutan gigi tikus putih (Rattus norvegicus).

English Abstract

The process of post-extraction sockets healing needs to be improved to prevent excessive alveolar bone resorption and restore alveolar bone function in supporting the teeth and protect important structures within. The leaves of tempuyung (Sonchus arvensis L.) contains triterpenoid. These compounds can inhibit osteoclastogenesis by inhibiting osteoclast-specific gene expression, namely Tartrate-resistance Acid Phosphatase (TRAP), so that osteoclast formation decreases and the process of healing sockets after tooth extraction will increase. The purpose of this study was to determine the effect of giving tempuyung (Sonchus arvensis L.) leaves extract on the number of osteoclasts in post tooth extraction sockets healing in white rats (Rattus norvegicus). This research was conducted by laboratory experimental method in vivo and divide the experimental animals into 4 groups, namely K7 and K14 (not given tempuyung leaves extract) and P7 and P14 (given tempuyung leaves extract), then decaputation was conducted on day 7 and 14. After that, histological preparations were made with HE staining and the amount of osteoclasts were counted. Data analysis using unpaired T-test showed significant differences in each of the 2 groups compared, except with K7 and P7. The Pearson correlation test showed a negative relations, which means that the longer tempuyung leaves extract were administered, the lower the number of osteoclasts will be. Based on this study, it can be concluded that the leaves of tempuyung (Sonchus Arvensis L.) affect the number of osteoclasts in healing process of post-tooth extraction sockets in white rats (Rattus norvegicus).
