Abstract
Plant growth promoting rhizobacteria adalah kelompok koloni bakteri di sekitar perakaran tanaman dengan berbagai peran yang menguntungkan bagi tanaman. Produktivitas padi di Kabupaten Merauke, Provinsi Papua yang masih rendah dengan luas wilayah potensial 614,9 ha memerlukan dukungan bakteri rhizosfer untuk meningkatkan kandungan unsur hara tanah. Tujuan penelitian ini adalah untuk mengisolasi, mengevaluasi, dan mengidentifikasi bakteri rhizosfer non-simbiotik penambat nitrogen yang unggul dalam memproduksi IAA dan melarutkan fosfat. Isolat dipilih berdasarkan karakter morfologi koloni yang mampu tumbuh dalam media selektif Azotobacter (Azotobacter Mannitol Agar) dan Azospirillum (N-free medium). Isolat yang didapatkan dari kedua tersebut dan memiliki kemampuan mengikat nitrogen sebanyak 11 isolat. Isolat-isolat tersebut diuji kemampuannya dalam memproduksi IAA dan melarutkan fosfat (isolat unggul diidentifikasi berdasarkan sekuen 16S rDNA). Isolat yang berpotensi tinggi memproduksi IAA adalah T7 dan S12, masing-masing sebesar 22,68 μg/mL dan 25,93 μg/mL sedangkan pelarut fosfat yang tinggi adalah isolat T7 dan S12, masing-masing sebesar 25,18 μg/mL dan 26,19 μg/mL dengan waktu inkubasi 72 jam. Kedua isolat (T7 dan S12) teridentifikasi sebagai Genus Myroides dengan nilai similaritas masing-masing adalah 94 % dan 84 % terhadap Myroides odoratimimus CCUG 39352. Kedua isolat berpotensi sebagai penghasil IAA dan pelarut fosfat yang efektif sehingga dapat diterapkan sebagai agen biofertilizer untuk meningkatkan produktivitas padi.
English Abstract
Plant growth promoting rhizobacteria is a group of bacterial colonies around plant roots with various benefits role for plants. Rice productivity in Merauke Regency, Papua Province which is catagorized as low with the potential area of 614.9 hectare needs a support of rhizoephere bacteria to improve the soil nutrients. This study was aimed to isolate, evaluate, and identify non-simbiotic nitrogen-fixing rhizobacteria which were superior as IAA producer and phosphate solubilizer. The isolates were selected based on morphological characterizations of colonies that grow in the selective media of Azotobacter (Azotobacter Mannitol Agar) and Azospirillum (N-free medium). The isolates obtained from both media with nitrogen-fixing ability were 11 isolates. These isolates were then tested for its ability in producing IAA and solubilizing phosphate. The best isolate was identified based on 16S rDNA sequences. The isolates with the high IAA producers were T7 and S12 by 22.68 μg/mL and 25.93 μg/mL, respectively. While the high phosphate solubilizer were also T7 and S12 by 25.18 μg/mL and 26.19 μg/mL, respectively. The incubation time for both ability was occured at 72 hours. The two isolates (T7 and S12) were identified as a member of Genus Myroides with similarity values of 94% and 84%, respectively, towards Myroides odoratimimus strain CCUG 39352. Although both isolates were not a member of genus Azotobacter dan Azospirillum, these isolates were potentially to be applied as biofertilizer agent for improving the rice productivity.
