Abstract
Sipermetrin merupakan golongan pestisida. WHO menyatakan sipermetrin menjadi sangat berbahaya dan berefek negatif terhadap kesehatan jika bahan makanan yang dikonsumsi mengandung residu sipermetrin. Sipermetrin dimetabolisme di hati menghasilkan toksin diantaranya hidroksil radikal, sianida dan aldehida yang dapat menghasilkan reactive oxygen species (ROS). ROS dapat menyebabkan stres oksidatif. Pada ovarium sipermetrin dapat menyebabkan apoptosis pada sel granulosa ovarium dan menurunkan kadar E2. Stres oksidatif dapat dicegah dengan pemberian antioksidan seperti ekstrak daun teh hijau. Manfaat ekstrak daun teh hijau terhadap penyembuhan banyak penyakit dikarenakan adanya senyawa polifenol yang didalamnya terdapat kandungan katekin yang tinggi. Penelitian ini bertujuan untuk membuktikan bahwa ekstrak daun teh hijau dapat meningkatkan jumlah sel granulosa dan kadar E2 serum tikus Wistar betina yang dipapar sipermetrin. Penelitian ini menggunakan desain true experimental dengan pendekatan post-test only with control group. Penelitian ini menggunakan hewan laboratorium yang dipelihara di Laboratorium Farmakologi Fakultas Kedokteran Universitas Brawijaya Malang Indonesia. Penelitian ini menggunakan tikus betina dan pengambilan sampel secara random. Semua perlakuan telah disetujui oleh Komisi Etik Universitas Brawijaya, Malang, Indonesia (No.15/EC/KEPK-S2/02/2018). Penelitian ini dilaksanakan pada bulan Februari sampai dengan Maret 2018. Tikus Betina (Rattus norvegicus) dipilih secara random dibagi menjadi 5 kelompok. Kelompok kontrol negatif (KN), kelompok yang diperi sipermetrin 20 mg/KgBB/hari (KP), dan kelompok yang diberi sipermetrin 20 mg/KgBB/hari dan diberi ekstrak daun teh hijau 7, 14, 28 mg/KgBB/hari (secara berurutan kelompok P1, P2, dan P3). Jumlah sel granulosa diberi pewarnaan dengan hematoxylin eosin (HE) dan dihitung dengan software HSel, kadar E2 serum diperiksa dengan ELISA ASSAY. Berdasarkan analisis statistik dengan One-Way ANOVA, hasil menunjukkan bahwa pemberian ekstrak daun teh hijau 7, 14, 28 mg/KgBB/hari secara signifikan (p<0,05) meningkatkan jumlah sel granulosa ovarium dan kadar E2 serum menyerupai KN. Pemberian ekstrak daun teh hijau dosis 28 mg/KgBB/hari menyebabkan peningkata tertinggi pada jumlah sel granulosa dan kadar E2 serum. Analisis korelasi dengan Pearson Correlation antara sel granulosa ovarium dengan kadar E2 serum tikus betina menghasilkan koefisien korelasi = 0,744 dan p = 0,000. Koefisien korelatisi tersebut menindikasikan bahwa ada hubungan yang positif dan kuat antara jumlah sel granulosa ovarium dengan kadar E2 serum. Artinya, semakin tinggi jumlah sel granulosa ovarium maka semakin tinggi pula kadar E2 serum. Sebaliknya semakin rendah jumlah sel granulosa ovarium maka semakin rendah pula kadar E2 serum. Kesimpulannya, ekstrak daun teh hijau pada dosis 28 mg/KgBB/hari mampu meningkatkan jumlah sel granulosa ovarium dan kadar E2 serum tikus Wistar betina yang dipapar sipermetrin.
English Abstract
Cypermethrin is a class of pesticides. According to WHO, Cypermethrin becomes very dangerous and affects health if that consumed by humans still contain residual Cypermethrin. Cypermethrin is metabolized in the liver decomposes into various toxic compounds such as radicals hydroxyl, cyanides and aldehydes resulting in reactive oxygen species (ROS). ROS causes stress oxidative. In the ovaries can make apoptosis in ovarian granulosa cells and decrease levels of E2. Oxidative stress can be prevented by giving exogenous antioxidants such as green tea leaves extract (Camellia sinensis). The benefits of green tea leaves extract to ward off various diseases due to the high content of polyphenols is a high catechin compound. This study proposes to demonstrate that green tea leaves extract can increase the number of granulosa cells and E2 serum levels on female rat exposed to Cypermethrin. This study was conducted using a true experimental with post-test only with control group design. This study used animal laboratory was taken care at the Pharmacology Laboratory, Faculty of Medicine, Universitas Brawijaya, Malang, Indonesia. This study used famale rat and the sample determination was done by random. All experiments were conducted according to the principles of guide for the care and use of laboratory animals in Indonesia and were approved by The Ethical Committee Universitas Brawijaya, Malang, Indonesia (No.15/EC/KEPK-S2/02/2018). These experiments were done from February to March 2018. Female rats (Rattus norvegicus) were randomly selected and divided into 5 groups, control group (KN), a group given only Cypermethrin 20 mg/kg body weight/day (KP), groups given Cypermethrin 20 mg/kg body weight/day and then treated with green tea leaves extract 7, 14, 28 mg/kg body weight/day (P1, P2, and P3 respectively). Treatment was given for 28 days. The number of granulosa cells was colored with hematoxylin eosin (HE) and counted by HSel software, E2 serum levels were determined via ELISA ASSAY. According to statistical analysis by One-Way ANOVA, treatment results with green tea leaves extract 7, 14, and 28 mg/kg body weight/day significantly (p < 0,05) increased the number of granulosa cells and E2 serum levels relative to KN. Administration of green tea leaves extract 28 mg/kg body weight/day caused the greatest increase in the number of granulosa cells and E2 serum levels. Analysis of Correlation by Pearson Correlation between of Granulosa Cells Ovarium with E2 Serum Levels of female rats had correlation coefficient = 0,744 and p = 0,000. The correlation coefficient indicated there was a positive and strong correlation between the number of granulosa cells and E2 serum levels. This meant that the higher of the number of granulosa cells that the higher the serum E2 level. Otherwise, the number of granulosa cells was lower then the E2 serum level was lower. In conclusion, green tea extract at a dose 28 mg/kg body weight/day is useful to increase the number of ovarian granulosa cells and E2 serum levels in female rats exposed by cypermethrin.
