Abstract
Minyak atsiri getah pohon kenari (Canarium indicum L) mempunyai aktivitas anti inflamasi dan antioksidan. Peradangan bronkus dan remodeling saluran napas merupakan faktor utama terjadinya asma. Minyak atsiri getah pohon kenari digunakan sebagai terapi pada hewan model tikus asma akibat paparan OVA dan Lipopolisakarida (LPS) dari bakteri gram negatif. Tujuan penelitian ini untuk mempelajari potensi minyak atsiri getah pohon kenari terhadap ekspresi tumor necrosis faktor (TNF-α), perubahan gambaran sel epitel bronkus, kadar MDA dan aktivitas protease pada hewan tikus model asma. Penelitian ini menggunakan lima kelompok tikus, yaitu kelompok kontrol negatif, kelompok asma, kelompok terapi dosis 25 mg/kg BB, 50 mg/kg BB dan 100 mg/kg BB. Data ekspresi TNF-α, aktivitas protease dan MDA dianalisis statistik ANOVA. Hasil penelitian menunjukan pemberian terapi minyak atsiri getah pohon kenari secara signifikan (P<0,05) menurunkan MDA sampai 54%, aktivitas protease sampai 46%, TNF-α sampai 67% dan memperbaiki histopatologi epitel bronkus dengan dosis terbaik adalah 100 mg/kg BB. Penelitian ini membuktikan bahwa minyak atsiri dari getah pohon kenari sebagai alternatif terapi asma.
English Abstract
Essential oils of canarium tree resin (Canarium Indicum L) have antiinflammatory and antioxidant activity. Inflammation of the bronchi and airway remodeling is a major factor in the occurrence of asthma. Essential oils of canarium tree resin may be used as therapy in asthma rats due to exposure to Ovalbumin and Lipopolysaccharide from gram-negative bacteria. The purpose of this study was to study the potential of essential oils of canarium tree resin on the expression of TNF-α, changes in bronchial epithelial cell features, MDA levels and protease activity in asthma rats. This study used five groups of rats, namely the negative control group, asthma group, therapy group dose of 25 mg/kg body weight, 50 mg/kg body weight and 100 mg/kg body weight. Data TNF-α expression, protease activity, and MDA level were analyzed by ANOVA. The results showed that essential oils of canarium tree resin therapy significantly (p<0.05) decreased MDA level up to 54%, protease activity up to 46%, TNF-α expression up to 67% and also improved bronchial epithelium histopathology. This study proves that essential oils of canarium tree resin can be effectively used as asthma therapy.
