Abstract
Kandidiasis adalah penyakit ginekologis akibat jamur yang paling umum terjadi pada manusia. Penggunaan antijamur seperti Fluconazole dilaporkan telah meningkatkan resistensi pada kandidiasis. Tumbuhan kenikir (cosmos caudatus Kunth.) memiliki kandungan fitokimia seperti flavonoid, saponin, alkaloid, tanin, polifenol dan minyak astiri. Senyawa Flavonoid telah terbukti digunakan sebagai antijamur pada Candida albicans. Penelitian menggunakan desain penelitian true experimental. Isolat Candida albicans diperoleh dari Laboratorium Mikrobiologi Universitas Brawijaya Malang dengan kode Candida albicans SV-1148. Isolate ditanam pada medium Sabouraud Dextrose Agar (SDA). Kelompok sampel dibagi menjadi kelompok kontrol negatif dan kelompok perlakuan dengan dosis konsentrasi ekstrak etanol daun kenikir 5%,10%, 20% 40%, kontrol positif fluconazole (60μg/ml) dan kombinasi (C. caudatus 20%+FCZ 30 μg/ml). Sampel dilakukan inkubasi selama 24 jam pada suhu 28oC. Kemudian dilakukan pengukuran masing-masing variabel permeabilitas membran menggunakan Flow cytometri dan kondensasi DNA menggunakan Immunfluorescence.
English Abstract
Candidiasis is a fungal gynecological disease that is most common in humans. The use of antifungals such as Fluconazole has been reported to increase resistance to candidiasis. The kenikir (cosmos caudatus Kunth.) Plant has phytochemical content such as flavonoids, saponins, alkaloids, tannins, polyphenols and essential oils. Flavonoids have been shown to be used as antifungals in Candida albicans. The study uses a true experimental research design. Candida albicans isolates were obtained from the Microbiology Laboratory of Universitas Brawijaya Malang with the Candida albicans code SV-1148. Isolate is grown on Sabouraud Dextrose Agar (SDA) medium. The sample group was divided into a negative control group and a treatment group with a dose of ethanol extract kenikir leaf 5%, 10%, 20% 40%, positive control fluconazole (60 μg / ml) and a combination (C. caudatus 20% + FCZ 30 μg / ml ). The samples were incubated for 24 hours at 28oC. Then measurements of each membrane permeability variable using Flow cytometry and DNA condensation using Immunfluorescence. This study produced ethanol extract of kenikir leaves to increase the percentage of permeability and condensation of DNA in the culture of Candida albicans.
