Abstract

Gentamicin merupakanantibiotikgolonganaminoglikosida yang bersifatnefrotoksik.Antibiotikinisecara normal tereliminasimelaluisistemurinasi.Namundalamdosis yang terlalutinggiakanmenumpuk di dalamlisosomepitel glomerulus danmengganggusistemfiltrasi glomerulus, zat-zat yang seharusnyadieliminasimelaluiurinakantertinggal di dalamalirandarah, salahsatunyaadalah BUN dankreatinin. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh gentamicin dalamdosisbertingkatterhadapfisiologiginjaldilihatdarikadar BUN dankreatinin di dalamdarah. Hewan model yang diujiadalahtikusputih (Rattusnorvegicus) jantan strain Wistardenganumur 2 bulandanberatbadan rata-rata 200 gram. Penelitian ini menggunakan empat kelompok tikus yaitu kelompok kontrol, kelompoktikusdengandosis gentamicin 30 mg/kgBB/hari, kelompoktikusdengandosis gentamicin 40 mg/kgBB/hari, dankelompoktikusdengandosis gentamicin 50 mg/kgBB/hari. Gentamicin diinjeksikanmelaluiintramuskularsebanyak 2 kali sehariselama 10 hari.Kadar BUN dan kreatinin diukur secaraujikimiadarah.Analisis data BUN dankreatinin yang digunakanadalahdenganmenggunakanuji ANOVA danujiTukey.Hasildaripenelitianiniadalah gentamicin dengandosis 30 mg/kgBB, 40 mg/kgBBdan 50 mg/kgBBtidakdapatmeningkatkankadar BUN ataupunkreatinin di dalamdarah.

English Abstract

Gentamicin is an Aminoglycoside type of antibiotic which has nephrotoxicity as its side effect. In normal state, this antibiotic will be eliminated through the urination system but in high dose, it will be accumulated inside the lysosome of glomerulus epitels. This accumulation will interfere the kidney’s filtration system. Some substances which are supposed to be eliminated through urine, are remained in the bloodstream such as BUN and creatinine.The purpose of this research was to determinethe effect of gentamicin to renal physiology based on BUN and creatininelevel in blood. The animal model was 2 months old, male white rats (Rattusnorvegicus)weighed 200 grams by average. This research used four groups of rats. The first group was thecontrol group without any treatment, the second one was a group of rats induced by gentamicin dose 30 mg/kgBW, the third one was group of rats induced by gentamicin dose 40 mg/kgBW, and the last one was a group of rats induced by gentamicin dose 50 mg/kgBW. Gentamicin was injected intramuscularly, twice a day for 10 days. BUN and creatinine level in blood were measured using blood chemistry test. The data of BUN and creatininewas analyzed by ANOVA and Tukey. The result of this experiment was gentamicin induced with dose 30 mg/kgBW, 40 mg/kgBW and 50 mg/kgBW could not elevate BUN and creatininelevel in blood
