Abstract

Inflamatory Bowel Disease (IBD) merupakan penyakit pada gastrointestinal kronis yang terjadi pada saluran pencernaan dan disebabkan oleh beberapa faktor seperti infeksi, agen parasit dan efek negatif obat-obatan NSAID (Non-Steroid Anti- Inflamatory Drugs) salah satunya adalah Indometasin. Penelitian ini bertujuan untuk mengetahui efektifitas ekstrak daun glodokan tiang (Polyathia longifolia) yang berasal dari Indonesia dan Filipina terhadap profil protein duodenum dan ekspresi EGF (Epidermal Growth Factor) pada tikus model IBD. Ekstrak etanol daun glodokan tiang (Polyathia longifolia) mengandung flavonoid yang dapat digunakan sebagai alternatif terapi kasus IBD. Hewan coba yang dipakai yaitu tikus (Rattus norvegicus) jantan berumur 8-12 minggu yang dibagi menjadi tiga kelompok yakni, kelompok 1 diinduksi sulfasalazine 20 mg/kgBB, kelompok 2 dan kelompok 3 diterapi dengan ekstrak etanol daun glodokan tiang (Polyathia longifolia) dari Indonesia dan dari Filipina dengan masing masing dosis 300 mg/kgBB, secara per oral selama 7 hari. Variabel yang diamati adalah profil pita protein diamati dengan metode SDS-PAGE yang dianalisa secara deskriptif semi kualitatif menggunakan Gel-doc dan ekspresi EGF pada duodenum dengan metode imunohistokimia yang dianalisis dengan ANOVA (Analysis of Variance) dan Uji BNJ ( Beda Nyata Jujur). Hasil penelitian menunjukan bahwa pada ketiga terapi mengekspresikan beberapa berat molekul protein sebesar 289,7 kDa, 45,6 kDa dan 21,2 kDa yang perlu dilanjutkan dengan metode western blot dan southern blot. Dan hasil perhitungan ekpresi EGF dari ketiga terapi mengalami perbedaan signifikan (P<0,5) antar kelompok sulfasalazine, ekstrak daun glodokan tiang (Polyathia longifolia) dari Indonesia dan Filipina.

English Abstract

Inflamatory Bowel Disease (IBD) is a chronic gastrointestinal disease that occurs in the digestive tract and is caused by several factors such as infection, parasitic agents and the negative effects of NSAIDs (Non-Steroid Anti-Inflammatory Drugs), one of which is Indomethacin. This study aimed to determine the effectiveness of glodokan tiang (Polyathia longifolia) leaf extract from Indonesia and the Philippines against the profile of duodenum protein and expression of EGF (Epidermal Growth Factor) in IBD model mice. Ethanol extract of glodokan tiang leaves (Polyathia longifolia) contains flavonoids which can be used as an alternative therapy for IBD cases. The experimental animals used were male rats (Rattus norvegicus) 8-12 weeks old divided into three groups, namely, group 1 was induced by sulfasalazine 20 mg / kgBB, group 2 and group 3 were treated with ethanol extract of glodokan tiang leaves (Polyathia longifolia) from Indonesia and from the Philippines with each dose of 300 mg / kg body weight, orally for 7 days. The variables observed were protein band profiles observed by the SDS-PAGE method analyzed semi-qualitatively using Gel-doc and EGF expression on the duodenum with immunohistochemical methods analyzed by Analysis of Variance and BNJ Test (Honest Real Difference). The results showed that on all three therapies expressed some protein molecular weight of 289.7 kDa, 45.6 kDa and 21.2 kDa which needed to be continued with the western blot and southern blot method. And the results of the EGF expression calculation of the three therapies experienced a significant difference (P <0.5) between the sulfasalazine group, glodokan tiang (Polyathia longifolia) leaf extract from Indonesia and the Philippines..
