Abstract

Hiperkolesterolemia merupakan kelebihan kolesterol di dalam darah. hiperkolesterolemia berkaitan erat dengan metabolisme lemak dan kolesterol yang terjadi di dalam hepar. Kerusakan hepar akibat hiperkolesterolemia dapat terjadi karena metabolisme kolesterol dan lemak sebagian besar terjadi di dalam hepar sehingga mempengaruhi fungsi hepar. Kerusakan hepar dapat ditentukan melalui pemeriksaan kadar SGPT (Serum Glutamic Piruvic Transaminase) dalam darah. Upaya pengendalian kadar kolesterol dapat menggunakan obat golongan statin, inhibitor absorbsi kolesterol, fibrat, asam nikotinat, namun dapat memberikan efek samping seperti gangguan hepar, gangguan saluran pencernaan, aritmia dan nyeri otot. Alternatif alami untuk meminimalisir efek samping dapat menggunakan bahan alami yaitu kulit pisang kepok. Kulit pisang kepok (Musa paradisiaca) mengandung senyawa pektin, flavonoid, tannin dan saponin yang berperan dalam menurunkan kadar kolesterol jahat di dalam tubuh. Penelitian ini bersifat True Experimental menggunakan Rancangan Acak Lengkap (RAL). Hewan model menggunakan tikus putih (Rattus norvegicus) jantan Strain Wistar berumur 8 – 12 minggu, dengan berat badan sekitar 100 – 150 gram diadaptasikan selama kurang lebih satu minggu dan dibagi dalam lima kelompok perlakuan yaitu kelompok negatif, kelompok positif, dan kelompok perlakuan 1, 2, 3. Pemberian diet hiperkolesterolemia menggunakan bahan asam kholat 0,1%, minyak babi 10%, dan kuning telur puyuh segar 5% dengan total pemberian 3,02 gr/2mL selama 21 hari. Pemberian ekstrak kulit pisang dengan dosis masing – masing 22,05 mg/150 g BB; 44,1 mg/150 g BB; 88,2 mg/150 g BB dan 44,1 mg/150 g BB dengan sonde lambung selama 28 hari. Analisis data terhadap kadar SGPT menggunakan uji One Way ANOVA dengan taraf kepercayaan 95% dan dilanjutkan uji Tukey menggunakan aplikasi SPSS 22.0. Gambaran histopatologi hepar dianalisa secara deskriptif. Hasil penelitian menunjukkan bahwa pemberian ekstrak kulit pisang kepok dapat mencegah kenaikan kadar SGPT pada dosis efektif 88,2 mg/150 gBB dan mampu mencegah terjadinya kerusakan sel hepar akibat pemberian diet hiperkolesterolemia.

English Abstract

Hypercholesterolemia is excess cholesterol in the blood. hypercholesterolemia is closely related to fat and cholesterol metabolism that occurs in the liver. Liver damage due to hypercholesterolemia can occur due to cholesterol metabolism and fat mostly occurs in the liver which affects liver function. Liver damage can be determined by examining SGPT (Serum Glutamic Piruvic Transaminase) levels in the blood. Efforts to control cholesterol levels can use statin drugs, cholesterol absorption inhibitors, fibrates, nicotinic acid, but can have side effects such as liver disorders, digestive tract disorders, arrhythmias and muscle pain. Natural alternative to minimize side effects can use natural ingredients namely kepok banana skin. Kepok banana skin (Musa paradisiaca) contains pectin compounds, flavonoids, tannins and saponins which play a role in reducing levels of bad cholesterol in the body. This study is True Experimental using a Completely Randomized Design (CRD). Model animals use male Wistar strain (Rattus norvegicus) white mice aged 8-12 weeks, weighing around 100-150 grams adapted for approximately one week and divided into five treatment groups namely negative group, positive group, and treatment group 1, 2, 3. Giving a diet of hypercholesterolemia using 0.1% kholat acid, 10% pork oil, and 5% fresh quail egg yolk with a total administration of 3.02 gr / 2 mL for 21 days. Provision of banana peel extract with each dose of 22.05 mg / 150 g BB; 44.1 mg / 150 g BB; 88.2 mg / 150 g BB and 44.1 mg / 150 g BB with a gastric sonde for 28 days. Data analysis on SGPT levels using the One Way ANOVA test with a confidence level of 95% and continued by the Tukey test using the SPSS 22.0 application. Hepatic histopathology description was analyzed descriptively. The results showed that administration of kepok banana peel extract could prevent an increase in SGPT levels at an effective dose of 88.2 mg / 150 gBB and was able to prevent the occurrence of liver cell damage due to the provision of a hypercholesterolemic diet.
