Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh zat pengatur tumbuh terhadap pertumbuhan akar adventif dan pengaruh elisitor logam berat Pb serta Cd terhadap pertumbuhan dan kandungan minyak atsiri vetiver oil pada akar adventif in vitro tanaman akar wangi. Akar adventif diinduksi dengan mengkulturkan tunas pada media MS + Kinetin (0,1 mg/L) + NAA (0; 0,5; 1; 3; 5 dan 7 mg/L). Akar adventif hasil induksi dikultur pada media MS + Kinetin 0,1 mg/L + NAA 7 mg/L + Pb atau Cd (0,1 mM) untuk elisitasi. Kultur diamati pertumbuhan dan dianalisis kandungan senyawa vetiver oil dengan GC-MS. Penambahan zat pengatur tumbuh NAA pada media mampu meningkatkan pembentukan akar adventif in vitro. Pertumbuhan akar adventif terbaik terdapat pada media MS + Kinetin 0,1 mg/L + NAA 7 mg/L. Penambahan elisitor logam berat Pb dan Cd berpengaruh terhadap pembentukan akar in vitro dan kandungan senyawa vetiver oil. Elisitor Pb meningkatkan jumlah akar, sedangkan elisitor Cd meningkatkan panjang akar. Tetapi penambahan elisitor Pb dan Cd pada media kultur menurunkan berat basah dan berat kering akar in vitro, . Akar adventif in vitro tanaman akar wangi teridentifikasi 58 senyawa vetiver oil. Senyawa utama vetiver oil pada akar tanaman akar wangi in vitro adalah Khusimone (6,94%), Khusimol (6,05%), Khusimene (4,85%), α Vetivone (3,70%), β Vetivone (3,53%), Vetiverol (3,22%), Prezizaene (2,35%), dan Zizaene (1,91%). Elisitasi dengan Pb dan Cd mampu meningkatkan persentase senyawa utama vetiver oil. Elisitor Cd meningkatkan persentase komponen senyawa utama vetiver oil lebih tinggi dibandingkan dengan elisitor Pb.

English Abstract

The objective of this research were to evaluate the effects of plant growth regulator on adventitious root growth and the effects of elicitors Pb and Cd on adventitious root growth and vetiver oil content of vetiver grass. Adventitious root were induced by culturing shoot explants on MS + Kinetin 0,1 mg/L + NAA (0; 0,5; 1; 3; 5 and 7 mg/L) medium. Adventitious root were cultured on MS + Kinetin 0,1 mg/L + NAA 7 mg/L + Pb or Cd (0,1 mM) medium for elicitation. Culture were observed, while vetiver oil content were analyzed by GC-MS. Increased plant growth regulator specifically NAA were able to increased adventitious root growth. The best root growth on MS medium contain Kinetin 0,1 mg/L + NAA 7 mg/L. Pb and Cd elicitor additions were influenced to root in vitro growth and vetiver oil content. 58 chemical compunds of vetiver oil were found on the adventitious root in vitro vetiver grass. Primary content of vetiver oil were Khusimone (6,94%), Khusimol (6,05%), Khusimene (4,85%), α Vetivone (3,70%), β Vetivone (3,53%), Vetiverol (3,22%), Prezizaene (2,35%), and Zizaene (1,91%). Elicitation with Pb and Cd were able to increased primary content of vetiver oil. Cd were better as elicitor on improving vetiver oil content than Pb.
