Abstract

Oksigen merupakan unsur vital dalam proses metabolisme seluruh sel tubuh. Paparan oksigen berlebih dapat menimbulkan Reactive Oxygen Species (ROS) atau radikal bebas yang menyebabkan stres oksidatif. Tujuan penelitian ini untuk mengetahui pengaruh pemberian ekstrak rosella ungu terhadap total bilirubin dan gambaran histopatologi organ ginjal pada mencit yang dipaparkan oksigen berlebih. Mencit yang digunakan dibagi menjadi 5 kelompok dengan perlakuan yang berbeda. Tiap kelompok, terkecuali kelompok pertama, diberi paparan oksigen berlebih sebanyak FiO2 0,8 selama 8 jam dalam waktu 7 hari. Kelompok pertama sebagai kontrol negatif (-) merupakan kelompok mencit sehat. Kelompok kedua sebagai kontrol positif (+) diberikan paparan oksigen berlebih tanpa diberikan terapi ekstrak rosella ungu. Kelompok 3,4, dan 5 adalah kelompok terapi yang berturut-turut dengan dosis 7,56 mg/20 g BB, 15,12 mg/20 g BB, dan 22,68 mg/20 g BB. Kadar total bilirubin diukur dengan spektrofotometri, dan gambaran histopatologi dilakukan pewarnaan HE. Data kadar total bilirubin dianalisis dengan uji One Way ANOVA dan Tukey Test dengan p<0,05, sedangkan untuk gambaran histopatologi dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa pemberian ekstrak rosella ungu pada setiap kelompok terapi menurunkan kadar total bilirubin, dimana dengan pemberian dosis 22,68 mg/20 g BB didapatkan hasil yang sama dengan kontrol negatif (p≥0,05). Hasil gambaran histopatologi dengan pemberian dosis 22,68 mg/ 20 g BB dapat mengembalikan struktur glomerulus seperti semula. Kesimpulan dari penelitian ini bahwa pemberian ekstrak rosella ungu dengan dosis 22,68 mg/20 g BB mampu menurunkan kadar total bilirubin dan memperbaiki gambaran histopatologi ginjal.

English Abstract

Oxygen is a vital element in the metabolic process of all body cells. Excessive oxygen exposure can cause Reactive Oxygen Species (ROS) or free radicals that cause oxidative stress. The purpose of this study was to determine the effect of purple rosella extract on total bilirubin and histopathology of kidney organs in mice exposed by excess oxygen. Mice used were divided into 5 groups with different treatments. Each group, with the exception of the first group, was given exposure to excess oxygen as much as FiO2 0.8 for 8 hours and carried out for 7 days. The first group as a negative (-) control was healthy mice. The second group as a positive (+) control was given excessive oxygen exposure without being given purple rosella extract therapy. Groups 3,4 and 5 were therapy groups with dose of 7.56 mg/20 g BW, 15.12 mg/20 g BW, and 22.68 mg/20 g BW, respectively. The total bilirubin level was measured by spectrophotometry, and histopathology was carried out by HE staining. The data of total bilirubin level was analyzed by the One Way ANOVA test followed by the Tukey Test with p<0,05, while the histopathological analysis was analyzed descriptively. The results showed that the administration of purple rosella extract in each treatment groups reduced the total bilirubin levels, which was given dose of 22,68 mg/ 20 g BW obtained the same results as a negative control (p≥0,05). The results of histopathology by given dose of 22,68 mg/20 g BW restore glomerular structure like a normal. In conclusion, the administration of purple rosella extract with dose of 22.68 mg / 20 g BW was able to reduce total bilirubin levels and improve kidney histopathology.
