Abstract

Common Snapping Turtle (Chelydra serpentina) adalah salah satu dari tiga spesies kura-kura famili Chelydridae. Common snapping turtle memiliki status least concern menurut International Union for Conservation of Nature (IUCN) dan special concern menurut Committe on The Status of Endangered Wildlife in Canada (COSEWIC). Penelitian ini bertujuan untuk mengetahui tingkat kekerabatan antar individu common snapping turtle dan membandingkan kedekatannya dengan sekuen gen CYT B alligator snapping turtle, central american river turtle, dan arizona mud turtle dari data NCBI. DNA didapatkan dari tiga sampel wholeblood common snappping turtle yang diisolasi menggunakan QIAamp® DNA Mini Kit. Pada penelitian ini, primer yang digunakan adalah (CYTB_F) 5’- GCGCCTCACTCTTCTTCATC-3’ untuk forward dan (CYTB_R) 5’- GCTGAGTTTAGGCCTGTTGG-3’ untuk reverse. Hasil dari PCR akan disekuensing menggunakan metode Sanger. Sekuen gen dianalisa menggunakan program Bioedit dan NCBI BLAST serta MEGA. Hasil penelitian menunjukkan bahwa jarak genetik antara sampel C1 Cekrik, C2 Mini, dan C3 Yeni terhadap database genebank Chelydra serpentina (accession number EF122793.1) sebesar 0%. Sedangkan jarak genetik ketiga sampel apabila dibandingkan dengan Macrochelys temminckii sebesar 13,9%, 20,8% dengan Dermatemys mawii, dan 23,5% dengan Kinosternon arizonense. Kesimpulan dari penelitian ini adalah sampel common snapping turtle (Chelydra serpentina) berkerabat dekat dalam satu spesies yang sama dan memiliki susunan sekuen DNA yang berbeda dengan Macrochelys temminckii, Dermatemys mawii, dan Kinosternon arizonense

English Abstract

The Common Snapping Turtle (Chelydra serpentina) is one of the three species of family Chelydridae. The status of Common snapping turtles is least concern according to International Union for Conservation of Nature (IUCN) and special concern according to Committe on The Status of Endangered Wildlife in Canada (COSEWIC). This study aims to determine the level of genetic relationships among common snapping turtle individuals and compared with the CYT B sequence of alligator snapping turtle, central american river turtle, and arizona mud turtle from NCBI data. DNA was obtained from two whole blood samples of common snapping turtle and isolated using QIAamp® DNA Mini Kit. The primers used in the PCR method consist of forward and reverse. In this study, we used (CYTB_F) 5'-GCGCCTCACTCTTCTTCATC-3’ for the forward primer and (CYTB_R) 5'-GCTGAGTTTAGGCCTGTTGG-3’ for the reverse primer. The results of PCR will be sequenced using the Sanger method. The gene sequence will be analyzed using the Bioedit, NCBI BLAST and MEGA programs. The research result shows the genetic distance between sample C1 Cekrik, C2 Mini, and C3 Yeni compared with database genebank of Chelydra serpentina (accession number EF122793.1) are 0%. Meanwhile, the genetic distance of the three samples compared to Macrochelys temminckii is 13,9%, 20,8% with Dermatemys mawii, and 23,5% with Kinosternon arizonense. The conclusion of this research is the samples of common snapping turtle (Chelydra serpentina) are closed related within one same species and has different DNA sequence arrangement between Macrochelys temminckii, Dermatemys mawii, and Kinosternon arizonense.
