Abstract
Masalah kesehatan yang sering terjadi pada kuda adalah robek tendon. Kerusakan tendon umumnya terjadi selama latihan. Inflamasi akan melepaskan sitokin IL-1β yang cenderung meningkat pada tahap awal cedera tendon serta adanya infiltrasi sel radang. Kelebihan produksi IL-1β terlibat dalam perubahan patofisiologis seperti arthritis, nyeri neuropatik, osteoarthritis, penyakit vaskular. Hal ini membutuhkan bahan yang dapat menurunkan IL-1β dan sel radang pada robek tendon. Getah nangka (Artocarpus heterophyllus) merupakan bahan alternatif yang mengandung pektin dan selulosa berfungsi sebagai perekat, menjaga kestabilan jaringan dan sel serta berperan sebagai antipiretik, dan antiinflamasi. Tujuan penelitian ini untuk mengetahui pengaruh getah nangka pada tikus (Rattus novergicus) model robek tendon terhadap ekpresi IL-1β dan jumlah sel radang. Pada penelitian ini digunakan tikus jantan dengan berat badan 150-200 gram berumur 2-3 bulan dibagi menjadi 4 kelompok. K- merupakan tikus sehat, K+ tikus robek tendon tanpa terapi, kelompok P1 tikus robek tendon dengan terapi Asam Mefenamat dan kelompok P2 tikus robek tendon dengan terapi Asam Mefenamat dan getah nangka sebanyak 10 mg/ekor. Robek tendon dilakukan dengan cara menginsisi daerah tendon achiles kaki kanan sepanjang 0,1 cm. Terapi dilakukan selama 14 hari. Ekspresi IL-1β dengan Imunohistokimia dan perhitungan jumlah sel radang dengan Pewarnaan haematoxyline Eosin merupakan data kuantitatif yang dianalisis menggunakan one-way ANOVA dengan uji lanjutan Tukey α < 0,05. Hasil analisa statistik jumlah sel radang dan ekspresi IL-1β menunjukkan bahwa kelompok P2 tidak berbeda nyata dengan kelompok tikus sehat (p>0,05), sehingga disimpulkan bahwa pemberian terapi getah nangka dapat menurunkan jumlah sel radang dan menurunkan sitokin IL-1β dalam penyembuhan robek tendon.
English Abstract
Health problems that often occur in horses are tendon rupture. Tendon damage generally occurs during exercise. Inflammation will release IL-1β cytokines which tend to increase in the early stages of tendon injury and the presence of inflammatory cell infiltration. Excess IL-1β production is involved in pathophysiological changes such as arthritis, neuropathic pain, osteoarthritis, vascular disease. This requires ingredients that can reduce IL-1β and inflammatory cells in tendon rupture. Jackfruit sap (Artocarpus heterophyllus) is an alternative material containing pectin and cellulose which functions as an adhesive, maintaining tissue and cell stability and acting as an antipyretic, and anti-inflammatory. The purpose of this study was to determine the effect of jackfruit sap on rats (Rattus novergicus) tendon rupture models on the expression of IL-1β and the number of inflammatory cells. In this study male rats with a weight of 150-200 grams aged 2-3 months were used divided into 4 groups. Kwas the healthy rats, K + was ruptured tendon rats without therapy, group P1was ruptured tendon rats with Mefenamic Acid therapy and group P2 was ruptured tendon rats with Mefenamic Acid therapy and 10 mg of jackfruit sap. Tearing of the tendon is done by insulating the area of the right foot achiles tendon as long as 0.1 cm. Therapy is carried out for 14 days. IL-1β expression with immunohistochemistry and calculation of the number of inflammatory cells with haematoxyline Eosin staining were quantitative data analyzed using one-way ANOVA with Tukey α <0.05. Statistical analysis of the number of inflammatory cells and IL-1β expression showed that P2 group was not significantly different from the healthy rat group (p> 0.05), so it was concluded that administration of jackfruit sap therapy could reduce the number of inflammatory cells and reduce IL-1β cytokines in healing tendon rupture.
