Abstract
Diabetes Mellitus (DM) adalah gangguan metabolisme yang disebabkan karena sel beta dalam pulau langerhans pankreas tidak dapatmenghasilkan insulin atau mengalami defisiensi insulin sehingga gula darahmeningkat (hiperglikemia). KondisiDM yang berlangsung lama dapat menyebabkan komplikasi ke berbagai organ akibat hiperglikemia yang muncul dan peningkatan radikal bebas. Pemberian ekstrak daun pegagan (Centella asiatica) diharapkan dapat mengurangi radikal bebas dan meningkatkan sensitivitas insulin. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak daun pegagan (C. asiatica) terhadap kadar Superoxide Dismutase (SOD) serum dan ekspresi Tumor Necrosis Factor Alpha (TNF-α) ginjal. Tikus (Rattus norvegicus) model DM dihasilkan dengan 40gram/ekor/hari pakan tinggi lemak selama 4 minggu dan pemberianmultiple low-dose Streptozotocin 30 mg/kg BB sebanyak dua kalidengan jarak 2 minggu secara intraperitoneal. Dua puluh ekor tikus dibagi menjadi lima kelompok tikus, yaitu Kontrol Negatif (kelompok sehat), Kontrol Positif (tikus DM),dan kelompok terapi P1, P2, P3 yang mendapat terapi dengan dosis 75, 150, dan 300 mg/kg BB ekstrak daun pegagan selama 1 minggu. Parameter yang digunakan adalah kadar SOD serum dengan uji spektofotometer dan ekspresi TNF-α ginjal dengan pewarnaanImunohistokimia (IHK). Analisis data SOD serum dan ekspresi TNF-α ginjal dengan uji One Way ANOVA dan uji lanjutan Tukey dengan tingkat kepercayaan 95% (α=0,05). Hasil penelitian menunjukkan bahwa terapi ekstrak daun pegagan (C. asiatica) dapat meningkatkan kadar SOD serum dan menurunkan ekspresi TNF-α ginjal secara signifikan (p<0,05) dengan dosis optimum 150 mg/kg BB. Kesimpulan dari penelitian ini yaitu esktrak daun pegagan (C. asiatica) dapat meningkatkan kadar SOD serum dan menurunkan ekspresi TNF-α ginjal pada tikus model DM.
English Abstract
Diabetes Mellitus (DM) is a metabolic disorder caused by beta cells in the island of langerhans failed to produce insulin,which lead to insulin deficiency and increasing of blood sugar level (hyperglycemia). Prolonged condition of DM can lead to various organs complications due to hyperglycemia and increased production of Reactive Oxygen Species (ROS). Centella asiatica leaf extract reduce ROS and improve insulin sensitivity. The object of this study were determine the effect of C. asiatica leaf extract on serum Superoxide dismutase (SOD) level and expression of kidney Tumor Necrosis Factor Alpha (TNF-α). Diabetic rats (Rattus norvegicus)prepared by inducing multiple low-dose streptozotocin of 30 mg/kg BW 2 times once a week and suplementation of hight fat diet dose 40 g/rat/day. Twenty rats were divided into five groups consist of Negative Control group (healthy rat), Positive Control group (Rat model of DM), P1, P2, and P3 with C. asiatica leaf extract dose therapeutic of 75, 150, and 300 mg/kg BW, respectively. Level of serum SOD analyzed by spectrophotometer and the expression of TNF-α of kidney observed by immunohistochemistry. Statistic analysis on serum SOD level and the expression of kidney TNF-α were using One Way ANOVA test continued with Tukey test at confidence level of 95% (α = 0,05). The results showed that treatment of C. asiaticaleaf extract significantly increased serum SOD levels and reduced kidney TNF-α expression (p <0,05) with 150 mg/kg BW as optimum dose. It can be concluded that the treatment withC. asiaticaleaf extract could increase Serum SOD levels and decrease kidney TNF-α expression on diabetic rats.
