Abstract
Inflammatory Bowel Disease (IBD) merupakan suatu penyakit pada saluran pencernaan diakibatkan adanya interaksi kompleks antara komponen imunoloigis, lingkungan, dan genetik. Inflammatory Bowel Disease (IBD) disebabkan oleh beberapa faktor, salah satu yaitu obat golongan Non Steroid Anti-Inflammatory Drug (NSAID). NSAID adalah obat yang sering digunakan sebagai obat analgesik, antiinflamasi, dan antipiuretik, yaitu indometasin. Daun sukun (Artocarpus altilis) memiliki kandungan flavonoid yang berfungsi sebagai antiinflamasi, antiplatelet, antioksidan, antimalarial, antimikroba, dan antikanker. Penelitian ini bertujuan untuk mengetahui pengaruh terapi ekstrak daun sukun (Artocarpus altilis) terhadap profil pita protein dan histopatologi kolon. Penelitian ini menggunakan tikus putih (Rattus novergicus) berkelamin jantan, berumur 8-12 minggu, dan dengan berat badan 150-200 gram. Terdapat 5 kelompok perlakuan, yaitu kontrol negatif, kontrol positif, kelompok terapi dengan dosis 100 mg/kg BB, 200 mg/kg BB, dan 400 mg/kg BB. Perhitungan profil pita protein dengan menggunakan metode Sodium Dedocyl Sulfate Polyacrylamid Gel Electrophoresis (SDS-PAGE) dan histopatologi kolon dengan menggunakan pewarnaan Hematoxylin-Eosin (HE). Hasil penelitian ini adalah ekstrak daun sukun dengan dosis 400 mg/kg BB dapat memperbaiki profil protein pada berat molekul 60 kDa dan 72 kDa serta mampu memperbaiki kerusakan mukosa kolon. Kesimpulan dari penelitian ini yaitu ekstrak daun sukun dapat digunakan sebagai terapi IBD
English Abstract
Inflammatory Bowel Disease (IBD) is a chronic inflammation condition of gastrointestinal tract caused by involve comlex interaction among immunologic, environmental, and genetic components. One of the factor of IBD is caused by NSAID. Non Steroid Anti-Inflamatory Drug (NSAID) is often used as an analgesic, anti-inflammatory and antipyuretic drugs, such as indometasin. Breadfruit leaves (Artocarpus altilis) have a content of flavonoid. Flavonoid have a function as antiinflammatory, anti-platelet, anti-oxidant, anti-malarial, anti-microbial, and anticancer. Thi study aims to determine the effect of breadfruit leaf extract against protein bands profile and colon histopathology. This study using male white rats (Rattus novergicus) weighing average 200 gram as experimental animals. This study using five groups are one for negative control group, one for positive control group, and three for therapy control groups which has each dosage 100 mg/kg BW, 200 mg/kg BW, and 400 mg/kg BW. Protein band profile is using Sodium Dedocyl Sulfate Polyacrylamid Gel Electrophoresis (SDS-PAGE) method and histopathology of colon is stained with Hematoxylin-Eosin (HE). The result of this study are a 400 mg/kg BB dose of breadfruit leaf extract can improves the protein profile at 60 kDa and 72 kDa and also is able to repair damage to the colon mucosa. The conclusion of this study is breadfruit leaf extract can be used as IBD therapy
