Abstract

Jumlah penderita kanker payudara terus mengalami peningkatan setiap tahunnya. Kaum wanita memiliki resiko lebih besar untuk terserang kanker ini. Pengobatan medis seperti kemoterapi sering menimbulkan efek samping bagi penderitanya. Penelitian ini dilakukan bertujuan untuk mengetahui pengaruh dari efek pemberian ekstrak kenikir (Cosmos caudatus) terhadap jumlah sel CD11b dan B220 pada mencit (Mus musculus) betina kanker payudara. Penelitian dilaksanakan pada bulan Januari - April 2019. Metode yang digunakan menggunakan Rancangan Acak Lengkap (RAL) yaitu mencit dibagi menjadi 5 kelompok, yaitu kelompok sehat (N), kelompok kanker (K), kelompok kanker yang diberi ekstrak kenikir 50 mg/kg BB (D1), kelompok kanker yang diberi ekstrak kenikir 500 mg/kg BB (D2) dan kelompok kanker yang diberi ekstrak kenikir 2000 mg/kg BB (D3). Hewan coba yang digunakan mencit betina strain BALB/c sebanyak 25 ekor berumur 8 minggu dengan berat badan 25-30 gram. Mencit diinduksi menggunakan DMBA dengan dosis 0,015 mg/gram, diinjeksikan secara subkutan selama minimal 6 minggu. Deteksi kanker dilakukan dengan pengamatan histologi jaringan payudara. Ekstrak kenikir diberikan secara oral selama 14 hari. Pada hari ke 15 organ spleen diisolasi, selanjutnya dilakukan pewarnaan antibodi. Sel kemudian dianalisis dengan flow cytometry menggunakan program BD CellquestTMPro. Data selanjutnya dianalisis menggunakan software SPSS 16 dengan uji ANOVA satu arah dengan nilai α=5%, dilanjutkan dengan uji Duncan. Pemberian ekstrak kenikir selama 14 hari mampu menurunkan jumlah sel CD11b+,. akan tetapi belum menunjukkan keefektifitasannya pada jumlah sel B220+.

English Abstract

The number of breast cancer sufferers continues to increase every year. Women have a greater risk of developing this cancer. Medical treatments such as chemotherapy often cause side effects for sufferers. This study was conducted to determine the effect of the effect of giving kenikir extract (Cosmos caudatus) to the number of CD11b+ and B220+ cells in female breast cancer mice (Mus musculus). The study was conducted in January - April 2019. The method used was Completely Randomized Design (CRD), namely mice divided into 5 groups, namely healthy group (N), cancer group (K), cancer group given 50 mg / kg BW kenikir extract (D1), the cancer group that was given 500 mg/kg BB (D2) kenikir extract and cancer group who were given 2000 mg/kg BW (D3) kenikir extract. The experimental animals used were strain BALB/c female mice as many 25 with 8 weeks old, a body weight of 25-30 grams. Mice were induced using DMBA at a dose of 0.015 mg / gram, injected subcutaneously for at least 6 weeks. Cancer detection is done by observing histology of breast tissue. Kenikir extract was given orally for 14 days. On the 15th day the spleen organs were isolated, then antibody staining was carried out. Cells were then analyzed by flow cytometry using the BD CellquestTMPro program. The data were then analyzed using SPSS 16.0 software with a one-way ANOVA test with a value of α = 95%, followed by the Duncan test. Giving kenikir extract for 14 days can reduce the number of CD11b+ cells. However, it has not shown its effectiveness in the number of cells B220+.
