Abstract

Inflammatory Bowel Disease (IBD) adalah penyakit peradangan pada bagian gastrointestinal tract (GIT) terutama kolon yang disebabkan karena efek pemberian Nonsteroidal Anti-Inflammatory Drugs (NSAID) seperti Indometasin dengan dosis pemberian 15 mg/kg BB per oral yang dapat menyebabkan inflamasi pada saluran cerna. Inflamasi pada saluran cerna akan menyebabkan peningkatan aktivitas protease sehingga menyebabkan kerusakan pada sel. Penelitian ini bertujuan untuk mengetahui potensi mucoprotectant dari larutan tepung tapioka yang memiliki kandungan resistant starch sehingga diharapkan mampu menurunkan aktivitas protease dan memperbaiki kerusakan pada histopatologi kolon tikus (Rattus norvegius) jantan usia 8-12 minggu dengan berat 150-200 gram. Tikus dibagi menjadi 5 perlakuan, yaitu kontrol negatif, kontrol positif, terapi 1 (0,9 mg/ 200 g BB), terapi 2 (1,8 mg/ 200 g BB), dan terapi 3 (3,6 mg/ 200 g BB). Terapi larutan tepung tapioka diberikan selama 14 hari. Pengaruh pemberian terapi diamati dengan pengamatan preparat histopatologi kolon dengan pewarnaan Hematoksilin-Eosin (HE) dan pengukuran aktivitas protease dari mukosa kolon. Analisa hasil perubahan kadar aktivitas enzim protease kolon antar kelompok dihitung menggunakan Analisa One Way ANOVA dan uji lanjutan Tukey atau BNJ (Beda Nyata Jujur) α<0,05 sedangkan analisa data histopatologi dari kolon dilakukan secara deskriptif kualitatif. Hasil penelitian menunjukkan bahwa pemberian terapi tepung tapioka mampu menurunkan aktivitas protease secara signifikan pada kelompok terapi 3 dengan dosis 3,6 mg/ 200 g BB (P3) sebesar 66,76% dan rata-rata aktivitas protease 1,24±0,04 µmol tirosin/g enzim menit serta mampu memperbaiki kerusakan pada mukosa kolon. Kesimpulan pada penelitian ini yaitu terapi larutan tepung tapioka dapat digunakan sebagai terapi alternatif IBD.

English Abstract

Inflammatory Bowel Disease (IBD) is an inflammatory disease of the gastrointestinal tract (GIT) especially the colon caused by the effects of giving Nonsteroidal Anti-Inflammatory Drugs (NSAIDs) such as Indomethacin at dose of 15 mg / kgBW orally. Indometachin would be given an impact of increasing the number of proteases so that protease activity increased and caused damage to colonic tissue. This study aimed to determine the potential of mucoprotectant from tapioca flour solution that had resistant starch so that it was expected to reduce protease activity and repair damage of rat colon based on histopathology. The rats (Rattus norvegicus) were aged 8-12 weeks with a weight of 150-200 grams. Rats were divided into 5 groups, namely negative control, positive control, therapy 1 (0,9 mg / 200 g BW), therapy 2 (1,8 mg / 200 g BW), and therapy 3 (3,6 mg / 200 g BW). Therapy for tapioca flour solution was given for 14 days, then rats were euthanized and the colon was taken for protease activity test and histopathological examination. The effect of therapy was observed by the colon histopathology preparations with Hematoxylin-Eosin (HE) staining and measuring protease activity from the colon. Analysis of the results of changed in levels of colon protease enzyme activity among groups would be analyzed statistically using One Way ANOVA and continued to Tukey test with α<0.05 and colon histopathology was analyzed qualitatively by tissue description. The results showed that tapioca flour therapy 3 with 3,6 mg/ 200 g BW dose could reduce 66,76% of protease activity (1,24±0,037 µmol tyrosine/g enzyme minutes) and also be able to repair damage of the colon mucosa. The conclusion of this observation is tapioca flour therapy can be used as alternative therapy for IBD.
