Abstract

Hiperkolesterolemia merupakan suatu keadaan dimana kondisi kolestrol melebihi batas normal di dalam tubuh yang ditandai dengan adanya peningkatan konsentrasi kolesterol serta Low Density Lipoprotein (LDL). Penelitian ini bertujuan untuk mengetahui pengaruh pemberian terapi angkak hasil fermentasi Monascus purpureus terhadap kadar kolesterol total dan gambaran histopatologi ginjal. Angkak merupakan hasil fermentasi beras yang dapat digunakan sebagai obat untuk hiperkolesterol yang memiliki kandungan senyawa statin yaitu berupa lovastatin. Lovastatin mampu menghambat Hydroxy-methyl-glutaryl Coenzyme A (HMG-CoA reduktase). Penelitian ini dilakukan bersifat eksperimental menggunakan Rancangan Acak Lengkap (RAL). Tikus putih (Rattus norvegicus) dibagi menjadi 5 kelompok, yaitu kelompok A (kontrol negatif), kelompok B (pemberian diet hiperkolestrol), kelompok perlakuan C (pakan diet hiperkolesterol dan angkak 0,5 gr/ekor/hari), kelompok perlakuan D (pakan diet hiperkolesterol dan angkak 1 gr/ekor/hari), dan kelompok perlakuan E (pakan diet hiperkolesterol dan angkak 1,5 gr/ekor/hari). Analisa data yang digunakan yaitu analisa kualitatif untuk gambaran hisopatologi ginjal dengan pewarnaan Hematoksilin-Eosin (HE) dan analisa kuantitatif untuk kadar kolesterol total dengan metode CHOD-PAP yang akan di uji statistik dengan one way ANOVA dan uji lanjutan yaitu uji tukey. Dilakukan pengamatan secara deskriptif pada gambaran histopatologi ginjal. Hasil penelitian menunjukkan bahwa terapi angkak dapat menurunkan kadar kolesterol total secara signifikan (P<0,05). Pengamatan histopatologi menunjukkan bahwa angkak dapat menghambat nekrosis dan mengurangi hipertrofi pada ginjal. Pada penelitian ini dosis terapi angkak 1,5 g/ekor/hari menunjukkan dosis terbaik dalam menurunkan kadar kolesterol total sebesar 50,51%. Dapat disimpulkan bahwa angkak dapat menurunkan kadar kolesterol total dan memperbaiki gambaran histopatologi ginjal tikus model hiperkolesterolemia.

English Abstract

Hypercholesterolemia is a condition where cholesterol exceeds the normal limits in the body which is characterized by an increase in cholesterol concentration and Low Density Lipoprotein (LDL). This study aims to determine the therapeutic effect of Monascus purpureus fermentation on total cholesterol levels and renal histopathological features. Angkak is the result of fermented rice which can be used as a drug for hypercholesterolemia which has a statin compound which is in the form of lovastatin. Lovastatin can inhibit Hydroxymethyl-glutaryl Coenzyme (HMG-CoA) reductase. This research was conducted in an experimental manner using a Completely Randomized Design (CRD). Rats (Rattus norvegicus) were divided into 5 groups, namely group A (negative control), group B (administered hypercholesterol diet), treatment group C (diet of hypercholesterolemia and angkak 0.5 gr/rat/day), treatment group D (diet of hypercholesterolemia and angkak 1 gr/rat/day), and treatment group E (diet of hypercholesterolemia and angkak 1.5 gr/rat /day). Data analysis used is qualitative analysis for the description of renal histopathology with Hematoxylin-Eosin staining (HE) and quantitative analysis for total cholesterol levels with the CHOD-PAP method which will be tested statistically with one way ANOVA and follow-up tests the tukey test. Descriptive observations were made on the histopathology of the kidneys. The results showed that Angkak therapy significantly reduced total cholesterol levels (P<0.05). Histopathological observations show that Angkak can inhibit necrosis of renal tubules, then reduce glomerural hypertrophy, improve kidney histopathology. A dose of 1.5 g/rat/day showed the best dose in reducing total cholesterol levels was by 50.51%. It can be concluded that red yeast rice can reduce total cholesterol levels and improve the histopathological picture of renal hypercholesterolemic rats.
